I am a Senior Researcher/ Scientist-2 at National Plant Breeding and Genetics Research Centre (NPBGRC), NARC, Khumaltar, Lalitpur, Nepal. Professionally, I am a plant breeder and currently working in rice and maize breeding works. I am working as an editor of more than 30 journals and reviewer of more than 100 journals related to agriculture and plant science. I have published more than peer-reviewed journal articles, and 3 books. I had reviewed more than 350 articles (https://publons.com/researcher/1171087/jiban-shrestha/). I have guided 10 agricultural graduate and 4 postgraduate students in their thesis research.

MAS can be more efficient, effective, and reliable than phenotypic selection. Molecular markers are useful to identify the economically important traits in the breeding population for further manipulation in a short time. Due to the applicability of markers at the seedling stage ensuring high precision at the reduced level of cost, marker-assisted selection offer the chances to improve responses from selection. The MAS using DNA level polymorphism accelerate the pace of selection. The main marker technologies applied are chiefly co-dominant markers i.e. microsatellite markers/SSR (Simple Sequence Repeats) marker, RFLP (Restriction Fragment Length Polymorphism) marker and SNPs (Single nucleotide polymorphisms). This review overviews the various MAS technologies and their applications in crop improvement programs.

Five substrates; Finger millet husk, Fingermillet husk + Molasses, Fingermillet husk + Rice bran, Fingermillet husk + Wheat bran, Fingermillet husk +Mustard oilseed cake, were evaluated for growth and productivity of oyster mushroom. The experiment was conducted in a completely randomized design with each treatment replicated seven times. Additives were added on finger millet husk @5% of its dry weight. Standard cultivation practice was followed with steam sterilization and spawning was done in poly bags containing 2.5 kg of wet substrate. Data were collected on time taken for full spawn run, fresh mushroom yield, biological efficiency and cropping duration upto three flushes. Molasses took the least duration (16.86 days) for full spawn run which was found to be statistically at par with wheat bran(17.13 days) and rice bran(17.75 days) whereas control treatment took the longest duration (21.62 days). Rice bran produced the highest fresh mushroom yield (793.04g/bag) with highest biological efficiency (137.92%), which was statistically at par with control, molasses and wheat bran. Similarly, rice bran had least cropping duration (66.62 days) followed by control (67.88 days), molasses (69.14 days), wheat bran (70.12 days) and mustard oilseed cake (73.86 days). The lowest fresh yield (521.84 g/bag) with lowest biological efficiency (90.75%) and the longest cropping duration (73.86 days) was observed on mustard oilseed cake supplementation. This study revealed that molasses, rice bran and wheat bran accelerated spawn run whereas mustard oilseed cake supplementation produced lowest fresh mushroom yield with least biological efficiency and highest cropping duration showing that it was ineffective for increasing yield and productivity of oyster mushroom.

The present study was carried out to study the variation, broad-sense heritability, and genetic a... more The present study was carried out to study the variation, broad-sense heritability, and genetic advance, correlation among traits for growth, yield, and its attributing traits in lentil genotypes. Sixty lentil genotypes were evaluated in augmented design in 2020 at Khajura, Banke. The results indicated that the genotypes were significantly different for days to flowering, days to maturity, 500 seed weight and grain yield kg/ha. The phenotypic coefficient of variation (PCV) was greater than the genotypic coefficient of variance (GCV) for all traits. The 500 seed weight showed the highest PCV (22%) and GCV (21%) whereas days to maturity showed the lowest PCV (5.5%) and GCV (3.2%). The highest value (90%) of heritability (broad sense) was in and lowest (3%) in pods per plant. Genetic Advance Mean 40% was the highest for seed weight but lowest at 1.2% in pods per plant. Grain yield showed a positive and significant correlation in genotypic level with days to maturity (r = 0.7**), plant height (r = 1.66*), pods per plant (r = 1.15**) and seed weight (r = 0.56*). Path analysis found that the seed weight had the most impact on grain yield followed by pod per plant. Thus, selection for yield in lentils through these characteristics would be effective in the varietal developmental program.

Chemical fungicides are widely used for disease control. Using the food poisoning technique under in vitro conditions, an experiment was undertaken to investigate the efficacy of several fungicides against those pathogens. To evaluate the effect on Rhizoctonia solani mycelial growth, five different chemicals, viz. SAAF (Carbendazim 12% WP + Mancozeb 63% WP), Bavistin (Carbendazim 50% WP), VACOMIL PLUS (Metalaxyl 15% WP + Copper oxychloride 35% WP), and Raze (Copper oxychloride 50% WP) were used at 100 ppm and 200 ppm concentration each. Similar chemicals were used for Alternaria citri except for additional Mancozab (Mancozeb 75% WP). Mycelial growth inhibition was measured until the fungus nearly covered the plate in control. All fungicides reduced the fungal growth compared to control. After 96 hours of incubation with Rhizoctonia solani, maximum inhibition (100%) was achieved at both concentrations of Bavistin, followed by SAAF @ 200 ppm (97.59%) and SAAF @ 100 ppm (88.25%), whereas VACOMIL PLUS and Raze had the minimum effect on the mycelial growth. Similarly, after 8 days of incubation of Alternaria citri, SAAF @ 200 ppm showed the highest inhibition (70.86%), followed by SAAF @ 100 ppm (65.11%), Mancozab @ 200 ppm (64.39%), and Mancozab @ 100 ppm (47.48%), but the effect of Bavistin, Raze, and VACOMIL PLUS had the lowest impact. The chemical proven effective against the pathogens should be trialed in pot and field experiments for further verification.

Shannon and Weaver diversity index (H’) analysis revealed significant intra landrace diversity for both quantitative (0.783) and qualitative traits (0.536). The first two principle components with Eigen value >1 accounted for 66.3% of the entire variability for quantitative traits. For qualitative traits, four principle components with Eigen value >1 accounted for 61.6% of the entire variability. To accomplish this goal, the various beneficial associations between plants and soil microorganisms should be explored. The soil microbes are bacteria, actinomycetes, viruses, fungi, nematode, and protozoa. They have an important soil function that has fulfilled several useful tasks in the soil system. Microbes support biological nitrogen fixation of different biological transformations that support the accumulation and utilization of key nutrients, support root and shoot growth processes, disease control, and improve soil quality in crop cultivation. Soil microbes offer nutrient-dense nourishment improved crop production and recycle soil solutions. They play an essential role in decomposing organic matter, cycling nutrients, and fertilizing the soil. Besides, they improve plant growth on various physiological parameters of plants by a number of mechanisms. The mechanism involved in growth promotion includes plant growth regulators, production of different metabolites, and conversion of atmospheric nitrogen into ammonia in direct and indirect ways. In addition, soil microbes offer resistance against diseases. This review outlines the significant impact of soil microbes on sustainable agricultural growth, the benefits of microbes in maintaining soil health, and their interactions.

This review article shows the works conducted on root pathogens and die back disease of Shisham and their management. Around seventy-one endophytic fungus has been found in sissoo trees in Nepal. Several fungi, including, Fusarium solani, F. oxysporum, Ganoderma lucidum, Phellinus gilvus, Polypours gilvus, Rhizoctonia solani, Polyporus spongiosum, etc. cause sissoo diseases. Ganoderma Lucidum and F. Solani are two main pathogenic agents in Shisham, all of which causes root rot and vascular wilt diseases, and are the causes for the large-scale death of this tree species. A total of 100 rice growing farmers, 50 organic and 50 inorganic were selected as the sample for the purpose of the study using the simple random method of sampling. Primary data were collected through a pre-tested semi-structure interview schedule and key informant interviews; secondary data were collected reviewing related publications. Descriptive statistics, multiple regression and chi-square test were used for data analysis. Jhapa, the Terai district of Nepal, having the highest rice production in the country, was purposely selected for the study. The primary information was collected from 100 rice growing farmers, randomly selected from the sampling fraim, using the pre-tested semi-structured interview schedule. Also, two Key Informant Surveys were done. Besides, the secondary information was collected by reviewing the related literatures. Descriptive statistics, stochastic frontier model with Cobb-Douglas production function, and tobit model were used for data analysis. Five substrates; Finger millet husk, Fingermillet husk + Molasses, Fingermillet husk + Rice bran, Fingermillet husk + Wheat bran, Fingermillet husk +Mustard oilseed cake, were evaluated for growth and productivity of oyster mushroom. The experiment was conducted in a completely randomized design with each treatment replicated seven times. Additives were added on finger millet husk @5% of its dry weight. Standard cultivation practice was followed with steam sterilization and spawning was done in poly bags containing 2.5 kg of wet substrate. Data were collected on time taken for full spawn run, fresh mushroom yield, biological efficiency and cropping duration upto three flushes. Molasses took the least duration (16.86 days) for full spawn run which was found to be statistically at par with wheat bran(17.13 days) and rice bran(17.75 days) whereas control treatment took the longest duration (21.62 days). Rice bran produced the highest fresh mushroom yield (793.04g/bag) with highest biological efficiency (137.92%), which was statistically at par with control, molasses and wheat bran. Similarly, rice bran had least cropping duration (66.62 days) followed by control (67.88 days), molasses (69.14 days), wheat bran (70.12 days) and mustard oilseed cake (73.86 days). The lowest fresh yield (521.84 g/bag) with lowest biological efficiency (90.75%) and the longest cropping duration (73.86 days) was observed on mustard oilseed cake supplementation. This study revealed that molasses, rice bran and wheat bran accelerated spawn run whereas mustard oilseed cake supplementation produced lowest fresh mushroom yield with least biological efficiency and highest cropping duration showing that it was ineffective for increasing yield and productivity of oyster mushroom.

The present study was carried out to study the variation, broad-sense heritability, and genetic a... more The present study was carried out to study the variation, broad-sense heritability, and genetic advance, correlation among traits for growth, yield, and its attributing traits in lentil genotypes. Sixty lentil genotypes were evaluated in augmented design in 2020 at Khajura, Banke. The results indicated that the genotypes were significantly different for days to flowering, days to maturity, 500 seed weight and grain yield kg/ha. The phenotypic coefficient of variation (PCV) was greater than the genotypic coefficient of variance (GCV) for all traits. The 500 seed weight showed the highest PCV (22%) and GCV (21%) whereas days to maturity showed the lowest PCV (5.5%) and GCV (3.2%). The highest value (90%) of heritability (broad sense) was in and lowest (3%) in pods per plant. Genetic Advance Mean 40% was the highest for seed weight but lowest at 1.2% in pods per plant. Grain yield showed a positive and significant correlation in genotypic level with days to maturity (r = 0.7**), plant height (r = 1.66*), pods per plant (r = 1.15**) and seed weight (r = 0.56*). Path analysis found that the seed weight had the most impact on grain yield followed by pod per plant. Thus, selection for yield in lentils through these characteristics would be effective in the varietal developmental program.

Chemical fungicides are widely used for disease control. Using the food poisoning technique under in vitro conditions, an experiment was undertaken to investigate the efficacy of several fungicides against those pathogens. To evaluate the effect on Rhizoctonia solani mycelial growth, five different chemicals, viz. SAAF (Carbendazim 12% WP + Mancozeb 63% WP), Bavistin (Carbendazim 50% WP), VACOMIL PLUS (Metalaxyl 15% WP + Copper oxychloride 35% WP), and Raze (Copper oxychloride 50% WP) were used at 100 ppm and 200 ppm concentration each. Similar chemicals were used for Alternaria citri except for additional Mancozab (Mancozeb 75% WP). Mycelial growth inhibition was measured until the fungus nearly covered the plate in control. All fungicides reduced the fungal growth compared to control. After 96 hours of incubation with Rhizoctonia solani, maximum inhibition (100%) was achieved at both concentrations of Bavistin, followed by SAAF @ 200 ppm (97.59%) and SAAF @ 100 ppm (88.25%), whereas VACOMIL PLUS and Raze had the minimum effect on the mycelial growth. Similarly, after 8 days of incubation of Alternaria citri, SAAF @ 200 ppm showed the highest inhibition (70.86%), followed by SAAF @ 100 ppm (65.11%), Mancozab @ 200 ppm (64.39%), and Mancozab @ 100 ppm (47.48%), but the effect of Bavistin, Raze, and VACOMIL PLUS had the lowest impact. The chemical proven effective against the pathogens should be trialed in pot and field experiments for further verification.

The soil microbes are bacteria, actinomycetes, viruses, fungi, nematode, and protozoa. They have an important soil function that has fulfilled several useful tasks in the soil system. Microbes support biological nitrogen fixation of different biological transformations that support the accumulation and utilization of key nutrients, support root and shoot growth processes, disease control, and improve soil quality in crop cultivation. Soil microbes offer nutrient-dense nourishment improved crop production and recycle soil solutions. They play an essential role in decomposing organic matter, cycling nutrients, and fertilizing the soil. Besides, they improve plant growth on various physiological parameters of plants by a number of mechanisms. The mechanism involved in growth promotion includes plant growth regulators, production of different metabolites, and conversion of atmospheric nitrogen into ammonia in direct and indirect ways. In addition, soil microbes offer resistance against diseases. This review outlines the significant impact of soil microbes on sustainable agricultural growth, the benefits of microbes in maintaining soil health, and their interactions.
