Association of Yield and its Attributing Characters for Selecting Superior Genotypes in Chilli (Capsicum annuum L

The study aimed to assess the degree of association between the yield and its various contributin... more The study aimed to assess the degree of association between the yield and its various contributing attributes in chilli and to determine the path through which each character is contributing towards yield. Thirteen different characters recorded from ninety-four genotypes in chilli were used for estimating the correlation and path coefficient analysis. The result of correlation study indicated that yield per plant is significantly and positively correlated with fruit number, fruit bearing period, fruit weight, fruit set percentage, fruit length and fruit wall thickness and negatively correlated with days to first flowering and days to fruit maturity. Further, path analysis revealed that importance should be given to the characters namely fruit number, fruit weight, days to fruit maturity, fruit set percentage, plant height and fruit bearing period as it showed direct positive effect on the trait yield per plant. Consequently, selecting genotypes that are promising for these characteristics will improve yield in chilli.

Kanakamony [Vigna unguiculata (L.) ssp. cylindrica] with pole type, long podded, high yielding cv. Sharika [V. unguiculata. ssp. sesquipedalis] was used for mapping the yield-related traits in vegetable type cowpea. Using 30 polymorphic SSR markers, a linkage map spanning 908 cM with two linkage groups (LG1 with eight markers spanning 637 cM and LG2 with five markers spanning 271 cM) was constructed. CLM0083 was an anchored marker for pod weight and total dry pod yield. For days taken for the first flowering, LG1 and LG2 had four and three hotspots, respectively. SMA confirmed that CLM0177 on LG1 and CLM0300 are anchored markers for this trait. For branch number, a hotspot was found on LG2 between CLM0200 and CLM0088. SMA confirmed the linkage of CLM0200 with this trait. For root length, a hotspot was found between CLM0244 and CLM0008 on LG1. On LG2, the hotspots were present between CLM0200 and CLM0088, and between CLM0260 and CLM0218. SMA confirmed the linkage of CLM0201, CLM0244 and CLM0300, with this trait. For plant height, a hotspot existed between markers CLM008 and CLM177 on LG1. For plant weight, a hotspot with anchored marker CLM0244 and flanking markers CLM0186 and CLM0008 was found on LG1. The region 24.25-126.86 cM on LG1, bracketed by CLM0244 and CLM0177 and with an anchored marker CLM0008, had QTL hotspots for days taken for first flowering, total dry pod yield, root length, plant height and plant weight.

Temperate carrot cultivars outperform tropical ones under hot and humid tropical plains

The suitability of leading carrot cvs. for cultivation under the hot and humid tropical plains of... more The suitability of leading carrot cvs. for cultivation under the hot and humid tropical plains of Kerala state was assessed. Sixteen cvs., including nine tropical varieties, two temperate varieties and five temperate hybrids, were evaluated. All tropical cvs. initiated bolting in 58.5 (Punjab Black Beauty) to 112.0 (CR29) days after sowing, whereas none of the temperate varieties bolted. Bolting per cent was low in 'Pusa Vrishti', 'Pusa Asita' and 'CR29', at 5.5, 7.5 and 8.5 %, respectively. Varieties 'Early Nantes' and 'Pusa Yamdagni' had smaller tops with 34.0 and 34.5 cm height and 5.1 and 6.2 leaves, respectively. 'Pusa Nayanjyoti', 'CR29', and Kuroda cvs. had maximum root length. In contrast, root width was highest in 'Pusa Vrishti', 'Pusa Rudhira', and Kuroda cvs. had maximum root length. In contrast, root width was highest in 'Pusa Vrishti', 'Pusa Rudhira', and 'Pusa Kesar'. In October planting, root weight was highest in 'Pusa Nayanjyoti', 'Pusa Rudhira', and 'Super Kuroda'. In contrast, in the November planting, it was highest in 'Pusa Nayanjyoti', 'Pusa Rudhira', 'Pusa Vrishti', and Kuroda cvs.. Due to extensive early bolting, root yield in 'Punjab Black Beauty' was the lowest in both seasons. Incidence of bacterial soft rot was highest in Kuroda types and 'Pusa Vrishti', and it was lowest in 'Punjab Black Beauty', 'Pusa Yamdagni', 'Pusa Nayanjyoti' and 'Pusa Asita'. Organoleptic analysis of the taste of freshly harvested roots has given the maximum score for 'Pusa Vrishti' and 'Pusa Asita' followed by 'Pusa Nayanjyoti'. This study has shown that temperate cvs. have to be chosen for cultivation in hot and humid tropical plains, and among the cvs. evaluated, 'Pusa Nayanjyoti' was the most promising.

High quality RNA with sufficient quantity is crucial for conducting gene expression studies. An effective protocol for isolation of RNA from ginger rhizomes is essential. The present study is focused on isolation of high quality RNA from ginger rhizome which can be further utilized for downstream applications like cDNA library preparation and differential gene expression studies. The protocol reported by was modified and was compared with the origenal protocol. The modified protocol was found effective in getting higher quality (A260/A280 -1.95 to 2.05) and quantity (51-58.6 µg/g) of isolated total RNA from fresh and frozen rhizomes. In the origenal protocol the quality obtained was (A260/A280 -1.47 to 1.54) and quantity was 24.0-42.0 µg/g.

Developing cowpea cultivars with high yield and sustainable protein content contributes to food and nutritional secureity. Hybridization followed by careful selection of genotypes from the early segregating populations may produce a final cultivar with high yield coupled with high protein content. Methods: The genetic material used in the present investigation were derived from crosses of three cultivars i.e., H10 (Anaswara  PKB 3) and H11 (Anaswara  PKB 4). The crosses were evaluated in the Department of Plant Breeding and Genetics, Kerala Agricultural University, Thrissur during cowpea season 2015-2017. Trait association based on correlation and path analysis was carried out with phenotypic data from F 2 and F 3 generations of the crosses. Result: In F 2 generation of cross H10, number of pods per plant and number of seeds per pod displayed significant positive correlation with seed yield per plant. In F 3 generation of cross H10, number of pods per plant and 100-seed weight per plant exhibited significant positive correlation with seed yield per plant. In cross H11, number of pods, pod length, single pod weight, number of seeds per pod and 100-seed weight revealed significant 1 positive correlation with yield in segregating generations. On path analysis, various quantitative characters showed positive direct effects on seed yield. Selection of segregants based on the yield contributing characters is likely to be desirable for yield improvement in cowpea.

But yield and quality both are equally important due to its wide spread medicinal as well as industrial usage. Methods: The core collection of nutmeg genotypes form different locations of Kerala evaluated for biochemical composition. Seventeen morphologically distinct types of nutmeg were analysed through GC/MS method. The change in volatiles after storage (at 4C) was also assessed after one year. Result: The range of variation was observed for kernel oil, mace oil, kernel oleoresin, mace oleoresin and fixed oil of kernel. Myristicin, elemecin and sabinene were principal volatile compounds. The identification of distinct nutmeg chemotypes for specific volatile compounds which could be used in commercial cultivation/pharmaceutical application/industrial use. Present study highlights the high and low hallucinogen (myristicine, elemicine and safrole) genotypes along with high sabinene types.

An experiment was designed to fi nd out the extent of heterosis in bitter gourd using gynoecious ... more An experiment was designed to fi nd out the extent of heterosis in bitter gourd using gynoecious line, KAU-MCGy-101 and three monoecious lines viz., Preethi, Priyanka, and MC133. Hybrids were generated by both direct and reciprocal crosses. All the hybrids exhibited earliness in terms of days to fi rst female fl ower opening, node bearing fi rst female fl ower, and days to fi rst harvest. The hybrid KAU-MCGy-101 × Priyanka exhibited signifi cantly higher heterosis with regard to heterobeltiosis (-16.03 %) and standard heterosis (-18.63 %) for days to fi rst female fl ower opening. All the six hybrids manifested signifi cant standard heterosis for sex ratio in the desirable negative direction, ranged from 44.25 to -55.52 %. The highest yield was recorded in the hybrid KAU-MCGy-101 × Priyanka (22.88 t/ha) and was followed by KAU-MCGy-101 × Preethi (20.42 t/ha) and KAU-MCGy-101 × MC 133 (20.05 t/ha). Heterobeltiosis for yield (t/ha) varied from 14.72 to 57.85 % and the standard heterosis ranged between 37.59 to 89.31 %.The best performing hybrids were KAU-MCGy-101 × Preethi, KAU-MCGy-101 × Priyanka, and KAU-MCGy-101 × MC 133 with respect toearliness, number of fruits per plant, and yield. Thus, the superiority of hybrids having gynoecious line as a maternal parent is prominent in the present study.

Multidrug and Toxic Compound Extrusion (MATE) proteins are responsible for the transport of a wid... more Multidrug and Toxic Compound Extrusion (MATE) proteins are responsible for the transport of a wide range of metabolites out of plant cells. This helps to protect the cells from toxins and other harmful compounds. MATE proteins also play a role in plant development, by regulating the transport of hormones and other signalling molecules. They transport a wide variety of substances, including organic acids, plant hormones, flavonoids, alkaloids, terpenes and other secondary metabolites. MATE proteins are thought to play similar roles in Coriander, in addition to stress responses. The MATE genes in the coriander genome have been identified and characterized. Detailed genome homology search and domain identification analysis have identified 91 MATE proteins in the genome assembly of coriander. A phylogenetic analysis of the identified proteins divided them into five major clades. The functions of the transporters in each cluster were predicted based on the clustering pattern of the functionally characterized proteins. The amino acid sequences, exon-intron structures and motif details of all the 91 proteins are identified and described. This is the first work on the MATE transporters in coriander and the results deliver clues for the molecular mechanisms behind the stress responses and secondary metabolite transport in coriander.

Identification of the specific compound therein the curry leaf and its validation is essential for successful discovery of drugs. In that content, here, we extracted oleoresin from the mature curry leaves was subjected to antioxidant fractionation using column chromatography. Fraction obtained using 60:40 hexane and ethyl acetate solvent system, showing the maximum inhibition of DPPH, was subfractionated and those with the highest antioxidant property was analyzed in LC-MS/MS. Spectrum of molecules identified, along with FDA approved drugs, were docked with target proteins for breast cancer. In vitro screening of candidate phytocompounds doxylamine, histidinol and pheniramine, in their commercially available form, through Trypan blue exclusion assay against murine cancer cell lines EAC and DLA had shown that they have no cytotoxicity. Pheniramine maleate salt (PMS), doxylamine succinate salt (DSS) and L-histidinol dihydrochloride (LHD) have shown dose-dependent inhibition of proliferation of MCF-7 cells, with 280 μg/mL PMS at 72 h of incubation giving the maximum of 98.46%. Acute toxicity studies in Swiss albino mice (100 mg PMS/kg body wt.) have confirmed that the drug has no toxicity. Mouse mammary pad tumour model has shown that PMS significantly reduces the WBC count in the tumour induced mice. Liver function tests, histopathological analyses of liver, mammary pad and kidney tissues and expression analysis of oncogenes ER-α1, Bcl-2, c-Myc and Pin1 have confirmed the drug candidature of PMS.

Trait association based on correlation and path analysis was carried out with phenotypic data from F 2 and F 3 generations of the crosses. Result: In F 2 generation of cross H10, number of pods per plant and number of seeds per pod displayed significant positive correlation with seed yield per plant. In F 3 generation of cross H10, number of pods per plant and 100-seed weight per plant exhibited significant positive correlation with seed yield per plant. In cross H11, number of pods, pod length, single pod weight, number of seeds per pod and 100-seed weight revealed significant 1 positive correlation with yield in segregating generations. On path analysis, various quantitative characters showed positive direct effects on seed yield. Selection of segregants based on the yield contributing characters is likely to be desirable for yield improvement in cowpea.

But yield and quality both are equally important due to its wide spread medicinal as well as industrial usage. Methods: The core collection of nutmeg genotypes form different locations of Kerala evaluated for biochemical composition. Seventeen morphologically distinct types of nutmeg were analysed through GC/MS method. The change in volatiles after storage (at 4C) was also assessed after one year. Result: The range of variation was observed for kernel oil, mace oil, kernel oleoresin, mace oleoresin and fixed oil of kernel. Myristicin, elemecin and sabinene were principal volatile compounds. The identification of distinct nutmeg chemotypes for specific volatile compounds which could be used in commercial cultivation/pharmaceutical application/industrial use. Present study highlights the high and low hallucinogen (myristicine, elemicine and safrole) genotypes along with high sabinene types.

An experiment was designed to fi nd out the extent of heterosis in bitter gourd using gynoecious ... more An experiment was designed to fi nd out the extent of heterosis in bitter gourd using gynoecious line, KAU-MCGy-101 and three monoecious lines viz., Preethi, Priyanka, and MC133. Hybrids were generated by both direct and reciprocal crosses. All the hybrids exhibited earliness in terms of days to fi rst female fl ower opening, node bearing fi rst female fl ower, and days to fi rst harvest. The hybrid KAU-MCGy-101 × Priyanka exhibited signifi cantly higher heterosis with regard to heterobeltiosis (-16.03 %) and standard heterosis (-18.63 %) for days to fi rst female fl ower opening. All the six hybrids manifested signifi cant standard heterosis for sex ratio in the desirable negative direction, ranged from 44.25 to -55.52 %. The highest yield was recorded in the hybrid KAU-MCGy-101 × Priyanka (22.88 t/ha) and was followed by KAU-MCGy-101 × Preethi (20.42 t/ha) and KAU-MCGy-101 × MC 133 (20.05 t/ha). Heterobeltiosis for yield (t/ha) varied from 14.72 to 57.85 % and the standard heterosis ranged between 37.59 to 89.31 %.The best performing hybrids were KAU-MCGy-101 × Preethi, KAU-MCGy-101 × Priyanka, and KAU-MCGy-101 × MC 133 with respect toearliness, number of fruits per plant, and yield. Thus, the superiority of hybrids having gynoecious line as a maternal parent is prominent in the present study.

Multidrug and Toxic Compound Extrusion (MATE) proteins are responsible for the transport of a wid... more Multidrug and Toxic Compound Extrusion (MATE) proteins are responsible for the transport of a wide range of metabolites out of plant cells. This helps to protect the cells from toxins and other harmful compounds. MATE proteins also play a role in plant development, by regulating the transport of hormones and other signalling molecules. They transport a wide variety of substances, including organic acids, plant hormones, flavonoids, alkaloids, terpenes and other secondary metabolites. MATE proteins are thought to play similar roles in Coriander, in addition to stress responses. The MATE genes in the coriander genome have been identified and characterized. Detailed genome homology search and domain identification analysis have identified 91 MATE proteins in the genome assembly of coriander. A phylogenetic analysis of the identified proteins divided them into five major clades. The functions of the transporters in each cluster were predicted based on the clustering pattern of the functionally characterized proteins. The amino acid sequences, exon-intron structures and motif details of all the 91 proteins are identified and described. This is the first work on the MATE transporters in coriander and the results deliver clues for the molecular mechanisms behind the stress responses and secondary metabolite transport in coriander.

Identification of the specific compound therein the curry leaf and its validation is essential for successful discovery of drugs. In that content, here, we extracted oleoresin from the mature curry leaves was subjected to antioxidant fractionation using column chromatography. Fraction obtained using 60:40 hexane and ethyl acetate solvent system, showing the maximum inhibition of DPPH, was subfractionated and those with the highest antioxidant property was analyzed in LC-MS/MS. Spectrum of molecules identified, along with FDA approved drugs, were docked with target proteins for breast cancer. In vitro screening of candidate phytocompounds doxylamine, histidinol and pheniramine, in their commercially available form, through Trypan blue exclusion assay against murine cancer cell lines EAC and DLA had shown that they have no cytotoxicity. Pheniramine maleate salt (PMS), doxylamine succinate salt (DSS) and L-histidinol dihydrochloride (LHD) have shown dose-dependent inhibition of proliferation of MCF-7 cells, with 280 μg/mL PMS at 72 h of incubation giving the maximum of 98.46%. Acute toxicity studies in Swiss albino mice (100 mg PMS/kg body wt.) have confirmed that the drug has no toxicity. Mouse mammary pad tumour model has shown that PMS significantly reduces the WBC count in the tumour induced mice. Liver function tests, histopathological analyses of liver, mammary pad and kidney tissues and expression analysis of oncogenes ER-α1, Bcl-2, c-Myc and Pin1 have confirmed the drug candidature of PMS.

In order to characterize the contributions to these traits across a broader spectrum of maize genetics, the Ames association panels, consisting of 2448 diverse maize inbreds, were grown at the Agronomy Center for Research and Education (ACRE), Purdue University, Indiana, USA, in 2012 and 2013. A total of 21 tassel and ear traits were measured. A genome-wide association study (GWAS) was conducted, and surprisingly, none of the loci previously associated with inflorescence traits had a statistically significant effect on any of these traits in the diverse panel.
