Procedure:
Part I: Preparation Media
1. Total media amount is 750mL which consist of 500mL of agar and 250mL of broth
2. Weigh all the reagent:
a. Tryptone 10g
b. Yeast extract 5g
c. NaCl 10g
d. Agar 7.5g
e. Water 750mL
3. Pour tryptone, yeast extract and NaCl together into 1000mL of bottle flask.
4. Add 750mL of water into 1000mL of bottle flask and swirl until it mixed.
5. Then pour 250mL into 250mL of bottle for broth and 500mL into 500mL of bottle for agar
6. For agar, add 7.5g of agar into 500mL of bottle.
7. Cut the aluminum foil and wrap on both the bottle.
8. Setting the autoclave machine on liquid.
9. Place both agar bottle and broth bottle into autoclave when autoclave was setting.
10. Remove the solution after finished and allow the agar to cool to 55C.
11. Keep the area sterile when pouring the plate by working near the flame or Bunsen burner.
12. Measure the agar 30mL for each plate by using sterile graduated cylinder.

Part II: Inoculation E.coli


1. Sterile working area by using 70 percent of Ethanol
2. Using flame or Bunsen burn while working on the inoculation E.coli.
3. Using 15 mL of centrifugal tube and approximately 2 mL of Broth to inoculate with BL21DE3
Competent E.coli
4. Then store in the incubator for 18 hours.
Part III: Preparation Medicine
Part I: Not fresh medicine
1. Weight the amount of each herb:

2. Label name and the amount of concentration on each 600mL of beaker.


3. Put each sample into each 600mL of beaker except Bai Zhu , Dang Zhen and Gan Jiang put in
the same beaker by following the table.
4. Add water into 600mL of beaker, follow the volume from the table.
5. Place on them on the hot plate then boiling for 40-60 minute.
6. Store all sample in the refrigerator for 1 week.
Part II: Fresh medicine

Label name and concentration on the 600mL of beaker.
Put the sample into each 600mL of beaker and mix Bai Zhu, Dang Zhen and Gan jiang in the
same beaker by following the table.
Place on the hot plate and boil them for 40-60 minute.

Discussion
The experiment was substantiate the traditional medicine with E.coli. The concentration was different in
each sample. Including three kind of herb, such as Huo Xiang, Jin Yin Hua and combination of Bai Zhu,
Dnag Shen and Gan Jiang. These were popular Chinese herb that used to prevent from diarrhea, nausea,
poor appetite and fever, it was similar infection from E.coli symptom. The quality of Chinese medicine
was freshness, it was important to patient.

According to research, the characteristic of Chinese medicine was a liquid and also required to take it
when medicine was warming. The experiment was concerned about fresh or not fresh medicine, it was
depend on the time that had taken. To compare freshness and not freshness, these were separated
extraction in two part. The first part would be keep for 1 week after completing. The other part would
be used immediately after extraction was finished. The concentration in fresh part would be comprise
two concentration, such as concentration one that was same as non-fresh concentration one too. And
the new concentration that dissimilar with other sample.
Throughout the experiment, we had two single and three combination herb. We changed the amount of
herb in every concentration and water. The volume of water specified from the highest weight of each
herb. In order to the concentration of each herb from greatest to smallest, the weight of Huo Xiang was
5g ,7.5g and 10g per 200 mL of water, Jin Yin Hua was 9g, 12g and 15g per 300mL of water and the last
one was three combination were 12g, 25g and 49g, all in fresh part. Non-fresh part, the new
concentration of Huo Xian , Jin Yin Hua and three combination increased serially to 18g and 21g, 15g and
30g, and 50g and 80g.
When the herb solution was completed, the process was ready for demonstration with E.coli. The
number of solution would be use about 110mml in each sample. This experiment would be test for five
time in each concentration, to examine precision of the solution with E.coli.
From the experiment, there were some error during the inoculation the agar, we changed some of
ingredient to Iodized refined salt, instead of NaCl because our laboratory did not have them. Secondly,
we begin with the non-fresh medicine. There were some of agar plate had been contract a disease that
was unusable for proving. As the result, all the sample cannot kill the bacteria then, we had a new
prediction from quality medicine had been reduced. That why we had the second part for support our
purpose.

Conclusion
We would see that all the medicine sample cannot kill the bacteria. We could prove that all these
sample were not use for patient who infect E.coli even the action was to prevent symptom that similar
as symptom of E.coli.
