D) Hanging-Drop Procedure

Materials:
Glass slides (glass slide with depression)
Plasticine
Coverslip
Micropipette and tips
Microscope
Bunsen burner
Young broth cultures of bacteria

Methods:
1. Obtain a clean coverslip by its edges and carefully put some plasticine on its
corners.
2. Place the cover slip on a paper towel, with the plasticine side up.
3. ransfer 30-40µL of the bacteria broth culture in the center of the prepared
coverslip.
4. Place a depression slide over the drop on the coverslip so that the plasticine
seals the coverslip to the slide.
5. Quickly invert it so the drop is suspended and the drop can be observed hanging
from the coverslip over the concavity.
6. Examine the drop under low power by locating the edge of the drop, reduce the
light with the iris diaphragm and focus.
7. Look through the eyepiece and raise the objective slowly using the coarse
adjustment knob until the edge of the drop is observed as an irregular line
crossing the field.
8. Observe the different sizes, shapes and typpes of movement.
*Please make sure differentiate between true motility and Brownian movement
9. When finished, discard the coversipls in the disinfectant jar and clean the slides.
