Subchronic Toxicty:
In a 5-day oral range-finding study, rats were administered daily doses of 50, 200 or 1000 mg/kg. Treatment-related
effects included increased liver weights, as well as erosion of the gastric mucosa and decreased food consumption
and body weight at the highest dose level. No histopathology was done. A marginal (non-significant) liver weight
increase was observed at the 50 mg/kg dose level which suggests that the no observable effect level (NOEL) may
be below this dose. Rats were treated orally with 5, 50, or 200 mg/kg/day for four weeks followed by a 2-week
recovery period. Microscopic findings included dose-related thyroid follicular hypertrophy. The lesions were milder
in females and, in the low dose group, males only were affected. They were essentially reversible. Increased liver
weights and cellular hypertrophy were seen in high-dose animals and were reversible. The NOEL was less than 5
mg/kg/day.
