Title TEST PROCEDURE For Determine the Total Fatty Matter

PURPOSE:

The purpose is to define the method of Total Fatty Matter

SCOPE:

This testing procedure will be applicable to Raw material, Finished Product.

PRINCIPLE OF METHOD
The emulsion is broken with dilute mineral acid and the fatty matter is extracted with petroleum ether . it
is weighted after removal of the solvent.

Requirements: Instruments /Solutions

1. Water Bath
2. Drying Oven
3. Dissector
4. Beaker / Flask
5. Separating funnel
6. Dry Sodium Sulphate
7. Dilute HCl – 1:1 (v/v)
8. Petroleum Ether(60-80C)
9. Hot Plate
10. Methyl orange indicator solution – Dissolve 0.1 Methyl orange in 100ml water.

PROCEDURE:

 Weight accurately 2 to 5gm. sample in a 250 ml conical flask .


 Then add 50 ml Dil. HCL fit a reflux condenser in to the flask and boil the contents until the solution is perfectly
Clear.
Pour the contents of flask in to separating funnel & allow it to cool to room temp, Rinse the conical flask with 50
ml petroleum ether in a portion of 10 ml.
 Pour the Petroleum ether rinsing in to the Sep. Funnel, Shake the Separating funnel well & leave until the layer
seprate.Seprate out the aqueous phase & shake it out with 50 ml portion of petroleum ether twice.
 Combine all ether extracts & wash with water until free of acid(When tested with methyl orange indicator)
 Filter the petroleum ether extract through filter paper containg sodium sulphate.
 After filtration dry at 90 +/- 2 deg.& then weight.

CALCULATION: Total Fatty Matter (%) = 100 X M1
