Microbiology

This document discusses methods for microbial control, including physical, chemical, and mechanical approaches. Physical methods use heat, radiation, or filtration to destroy microbes. Heat methods include incineration, dry heat, steam, and boiling water. Radiation includes ionizing sources like X-rays and gamma rays, and nonionizing sources like UV light. Chemical agents include gases and liquids that can be used for sterilization, disinfection, or antisepsis. Mechanical removal methods use filtration of air or liquids. The effects of irradiation on microbes and applications of irradiation control are also covered.

MICROBIOLOGY

GENERAL CONSIDERATIONS IN MICROBIAL CONTROL

Physical, chemical, and mechanical methods to destroy or reduce undesirable microbes in a given
area (decontamination)

Primary targets is microorganism capable of infection of spoilage

 Vegetative bacterial cells and endospores
 Fungal hyphae and spores, yeast
 Protozoan trophozoites and cysts
 Worms
 Viruses
 Prions

MICROBIAL METHOD

PHYSICAL AGENT

CATEGORY TYPES EXAMPLE ACHIEVE

Incineration sterilization
Heat DRY Dry oven Sterilization
Steam under pressure Sterilization
Moist Boiling water, hot Disinfection
water,
Ionizing . X-ray, cathode, Sterilization
Radiation gamma
Nonionizing UV Disinfection

MECHANICAL REMOVAL METHOD

CATEGORY TYPES ACHIEVE

FILTRATION Air Disinfection
Liquids Sterilization

CHEMICAL AGENTS
CATEGORY TYPES ACHIEVE
Gases Sterilization
Disinfection
Liquid Animate Antisepsis
Inanimate Disinfection and sterilization

Note:
 Disinfection: The destruction or removal of vegetative pathogens but not bacterial
endospores. Usually used only on inanimate objects.
 Sterilization: The complete removal or destruction of all viable microorganisms.
Used on inanimate objects.
 Antisepsis: Chemical applied to body surfaces to destroy or inhibit vegetative
pathogens.

CELLULAR EFFECTS OF IRRADIATION

1. Ionizing radiation - deep penetrating power sufficient
energy to cause electrons to leave their orbit
 Gamma rays, X rays, cathode rays
 Breaks DNA

2. Nonionizing radiation - little penetrating power

 UV light creates pyrimidine dimers
 Interferes with replication

APPLICATION OF IRRADIATION CONTROL
