Danger! Electric shock.


→ Switch off the device and disconnect the power plug before opening the device, performing
work on electrical connections or starting maintenance or cleaning work.

Risk of explosion!!
→ Do not operate the device in rooms where work is being carried out with explosive substances.
→ Do not use this device to process any explosive, radioactive or highly reactive substances.

Risk when handling toxic or radioactively-marked liquids or pathogenic germs.


→ Follow national regulations governing the handling of these substances.
→ For complete instructions regarding the handling of germs or biological material of risk group II
or higher, please refer to the "Laboratory Biosafety Manual" (Source: World Health Organization,
current edition of the Laboratory Biosafety Manual).

Warning! Risk of injury from sharp-edged components.


→ If the Mastercycler realplex module is propped open,sharp-edged components are accessible.
When the module is being cleaned, the sharp edges of the Rollflex cable and the guide rail may
lead to injuries.

Warning! Burns from hot thermoblock and heated lid.


→ The thermoblock and frame with heating foil very rapidly reach temperatures of 95°C respec-
tively 105 °C.
→ Risk of burns!
→ Keep realplex-Modul closed until the temperature of the thermoblock is below 30 °C.

Warning! Risk of burns.


→ The thermoblock, frame with heating foil and micro test tubes/PCR plates very rapidly reach
temperatures of above 105 °C.
→ Allow the block and the frame with heating foil to cool down to 30 °C before opening the device.
→ Symbol on thermoblock: warns that the thermoblock may be hot (triangle pointing downwards)
or that the frame with heating foil may be hot (triangle pointing upwards). This Symbol
shows that during gradient operation, the temperature on the right-hand
side is higher than on the left-hand side.

Warning! Injury from trapping or crushing.


→ Do not place your fingers between the lid and the housing when opening and closing the device
or when opening it into the cleaning position.
→ Do not reach in under the open lid.
→ Ensure that the prop is correctly engaged so that the device remains in its open position.

Caution! Damage to electronic components and the lens from condensation.


After moving the Mastercycler ep realplex from a cool environment to a warmer one, condensation
may form in the device.
→ Allow the Mastercycler ep realplex to warm up for at least 12 hours after being moved before you
connect it to the mains power supply.

Caution! Material damage from incorrect connections.


→ Only electrical connections to devices described in the operating manual may be made.
→ Other connections are permitted only following consultation and agreement with Eppendorf AG!

Warning! Damage due to incorrect power supply.


→ Only connect the device to power sources that match the electrical specifications on the device
ID plate.
→ Use only sockets with protective earth.

Caution! Damage due to overheating.


→ Do not place the device close to sources of heat (e.g. radiator, drying cabinet).
→ Do not expose the device to direct sunlight.
→ Ensure free circulation of air by maintaining a distance of at least 10 cm on all sides of the device
from adjacent devices or walls and keep the underside of the device clear.

Caution! Damage to the device from incorrect operation.


→ Do not put any objects down on the realplex-Modul. Do not allow any liquid to get into the
device.

Caution! Damage to the thermoblock from incorrect operation.


→ Do not pour sample material straight into the thermoblock.

Caution! Damage to the thermoblock from the use of unsuitable sample tubes.

1 Safety
Unsuitable tubes may be so severely damaged that sample material is released. Particular attention
should be paid to this point when working with infectious material.
→ Use only sample tubes recommended by Eppendorf.

Warning! Risk to health if used incorrectly.


If incubation is performed incorrectly, sample material may be released and the thermoblock and the
realplex-Modul damaged.
→ Ensure that the PCR plates are correctly inserted in the thermoblock.
→ Always heat the thermoblock and the frame with heating foil simultaneously. If incubation is per-
formed with the frame with heating foil cold, micro test tubes may burst.

Warning! Poor safety due to incorrect accessories.


The use of accessories and spare parts other than those recommended by Eppendorf may impair
the safety, function and precision of the device. Eppendorf accepts no warranty or liability for
damage caused by third-party parts or incorrect use.
→ Use only original accessories recommended by Eppendorf.

Caution! Damage to the display from mechanical pressure.


→ Do not exert any mechanical pressure on the display.

Caution! Ensure that service manuals are up to date.


→ Before you start working with the device make sure that your service manual is up to date. To do
so, check the version number at www.eppendorf-support.com.

The safety regulations applicable in the lab should always be observed when installing and using the
device.

Due to its weight of 24 kg, the Mastercycler ep realplex must be positioned completely on a stable
work surface. A secure stand is required when opening and closing the Mastercycler ep realplex.

Danger! Electric shock as a result of penetration of liquid.


→ Switch off the device and disconnect it from the power supply before starting cleaning or disin-
fecting.
→ Do not allow any liquids to penetrate the inside of the housing.
→ Do not disinfect by means of spraying.
→ Only reconnect the device to the power supply once it is completely dry.

A new PCB Power supply unit (6321 856.509) is available for Mastercycler pro 6321 / 6325
Mastercycler ep 5341 / 5344 / 5345 and Mastercycler realplex 6300 / 6302.

The new PCB Power supply unit (6321 856.509) replaces the previous PCB Power supply 6321 856.002
and 5341 856.002.

We recommend that you include this information together with Chapter 2 of your Mastercycler Service
Manual, to have all relevant product information available in one place.

For PCB Power supply unit (6321 856.002) a modified PCB SMPS ( 6321 895.008 ) is available.

For Mastercycler pro with PCB Power supply 6321 856.002 the PCB SMPS
(switch mode power supply) must be exchanged, if the errors D2C and/or D2D occasionally occur.

For Mastercycler ep 5341 / 5344 / 5345 and Mastercycler realplex 6300 / 6302
the previous PCB Power supply 6321 856.002 as well as the PCB Power supply 5341 856.002
are further on usable without any modifications.

We recommend that you include this information together with Chapter 2 of your Mastercycler Service
Manual, to have all relevant product information available in one place.

If an error occurred, a screenshot of the error message and the OdEvent.log will be needed for troubleshooting. Save
OdEvent.log before restarting the Mastercycler ep realplex system. This is the pathfinder C:/ Programs/Eppendorf/
realplex/OdEvent.log.If necessary system can be restarted now.

Differentiating feature of service functions within realplex software and commissioning software.
The service functions for the realplex module/Mastercycler ep are in the realplex software. These functions can be used
for testing.
The commissioning software needs to be installed on the realplex computer separately.
It is required only in the event of the realplex being serviced or repaired, for example for switching voltage or after
replacing a PCB or a multiplier.

realplex Application Software V1.5: Call up the service functions of realplex or Mastercycler.

Conditions
Connect the computer to the Mastercycler ep realplex via the USB port, the USB CAN adapter.

– Exit all other programs.


– Only start the realplex program now.
– Switch on Mastercycler ep realplex-system.
– Select "Service" as the user name and confirm with Enter.
– Enter the password "23456789" and confirm with Enter.

System Info
System information is adopted on commissioning
and written to the appropriate fields.
The possible range for "Current PMT Voltage Chn." is
between 0 and 255. Each channel can be tested in
stages 0 – 9.

Filterwheels
This function can be used to perform a complete filter
drum self-test.
A 360° rotation of the filter drum is 115,200
microsteps. The offset means the deviation from the
zero position.
Each filter has its own code.
2 = 605 nm
4 = dark filter
5 = 550 nm
10 = 580 nm
15 = 520 nm
The two slide controllers at the bottom of the function
can be used to rotate the relevant filter drum into any
desired position.
If a dot can be seen after "Light Barrier", there is a
coding initiator switch for a filter in the light barrier.

LED-Array
The "Flash" button can be used to sub-ject all the
LEDs on the PCB LED Matrix to a visual inspection.
The wavelength of the LEDs is 470 nm. It is not
necessary to wear safety glasses.
With the self-test, all LEDs are tested and the result
written into the table.
0 means OK.
A red cross means that the LED in the relevant
position is defective. If one LED is defective, the PCB
LED Matrix will have to be replaced.
A repair is then performed with the aid of the
commissioning software, followed by dye calibration.

Multiplier
A semi-skirted/black plate from test box 6302

875.004 is inserted for the multiplier test.


For this dark current measurement, multiplier voltage
is first switched off (Voltage off) and then on (Voltage
on). On a multiplier which measures in the "blue
range" (520 – 550 nm), the difference between
"Voltage off" and "Volt-age on" is very small.
"Voltage on" is always slightly higher. On a multiplier
which measures in the "red range" (580 – 605), the
"Voltage on" value is much higher because it
includes the effect of heat in the infrared range. If the
upward limit value is exceeded, there is an error
message.

Optical Path

function can be used to test the relevant channels.
Each channel can be tested individually using
"Channel & Filter Selection".

with figures.
0 – 4 is OK.
From 5, the range is exceeded and a red 5 appears.
If a yellow 5 can be seen, this means that a fault has
been found at this position on another channel.

Calibration
In order to verify calibration/ adjustment, the
Lumogen reference plate from test box

6302 875.004 needs to be inserted.
Under "defective wells" is a display of how many
positions are too low and how many are too high.
Too low means that the relevant light conductor is
defective or there is contamination of a lens array in
the light path. Too high means that there is contami-
nation. The mean values are shown under "mean".
Percentage details shown under "(dev.)" have been
calculated from the deviation of the actually
measured values to the stored calibration data.
Deviation may not exceed ±10 %.

Well State
For the Well State test, the Lumogen reference plate
from test box 6302 875.004 must be inserted.
Each channel can be tested individually using
"Channel & Filter Selection".
All 96 measuring positions are shown in the matrix.
In principle, this function does the same as
calibration, just showing it differently.

The commissioning software (IBSW) is a service test software available to download at www.eppendorf-support.com.
This commissioning software is required to adjust the realplex module.
Commissioning software version 1.14 requires Firmware V 3.36 on the PCB control.
– Connect the computer to the Cycler CAN input on the rear of the Cycler using the USB CAN adapter and the
PC–CAN bus cable.
– Connect the patch cable of Cycler CAN_out to realplex module CAN_in.

– Set the address of the Cycler to 02 using the address switch.


– Set the address of the realplex module to 11.
– Set the term switch on the Cycler in up position and on the realplex module to the down position.

– When carrying out an adjustment on the realplex system, the dark filter must be replaced with a pale filter before the
device is switched on in the filter wheel.
– To do this, remove the gray cover on the realplex module.
– Unscrew both screws and remove the cover.
– Replace the dark filter with the pale filter, replace the gray cover and tighten the two screws again.
– Plug the power supply cable into the computer and supply it with power.
– Switch on the computer.
– Switch on the realplex system.
– Install the Commissioning software.
– Start the Commissioning software.
 Note: only one application may ever run at a time.

Replacement of the PCB Control

The workplace must comply with ESD guidelines and have low dust levels.
This description applies to the RealPlex software V 1.5. This software version includes the firmware for the RealPlex
module 3.36 and for the Mastercycler module 2.601 or higher. The startup software (IBSW) is version 1.14.
If replacement of the PCB Control is necessary in case of service, all configuration data such as Device Nr., PCB versions
and multiplier types have to be re-entered. Adjustment with the IBSW is also required.

Procedure prior to installation of the PCB Control

– Synchronize system using IBSW to write all configuration data from PCB Control to IBSW.
– Then press key "Disconnect" so that the text file 630xxxxXXX_log_inf.txt is written. This file can be found stored
under C:/Programme/Eppendorf/IBSW.
– Exit IBSW.

Procedure after replacement of PCB Control

– Set address to 11.

– Set First Dev.-Nr. to 11.
– Press "Browse" key and select file "onlinedetection.bin".
– Press Update button to reflash PCB Control.
– Then restart IBSW and synchronize system.
– Open text file 630xxxxXXX_log_inf.txt, input configuration data manually and transfer to new PCB using
"Set data" function.
– Then perform adjustment.

Firmware update of PCB Control

If the PCB Control firmware is to be updated, this is possible using Start/Programme/Eppendorf/Firmware Update.
The firmware Update Tool shows the current firmware versions.
In this case (see below) no update is required.

– Pull back the ribbon cable to the opening in the baseplate.


– Fix the ground plate in place with at least two screws.
– Stand the module back on the slide bars.
– Unscrew the 4x T10 Torx screws to remove the upper bridge with the fan.
– Detach the four large springs.
– Unscrew the four T10 Torx screws from the LED Matrix component assembly.
– Lift off the LED Matrix component assembly. The assembly is guided with two pins.

– Lift the ground plate and remove plugs X7 and X8 for the multiplier from the PCB Control.
– Under the PCB Control there are the fastening screws for the relevant channel an with a two-channel device, also the
screw for the beam splitter. Unscrew the 2x T20 Torx screws for the corresponding channel, see Fig. 11.

– Fold down the ground plate and fix it in place with at least two screws.
– Stand the module back on the slide bars.

– Cut open relevant tie wrap.

– Multiplier spring remove 1x T20 Torx screw with washer. The multiplier spring is no longer necessary when a new
multiplier module is to be used. However, the T20 Torx screw for the spring is once again screwed into the threads in
order to prevent the incidence of stray light.

– Unscrew one of the two screws for the relevant power supply unit of the multiplier; the other one should just be
loosened.
– Remove the cover of the Amplifier housing.
– Unsolder the relevant multiplier cable on the PCB Amplifier.
– Unscrew the seven slotted screws of the cover plate.
– Lift of cover plate.
– Place the realplex module safely to one side.
– Pull the cable for the light barrier and the filter wheel motor through the cable bushing in the direction of the multiplier
module.
– The following parts from the old multiplier module are mounted on the new module:
First the filter wheel. To this purpose loosen the M2 socket-head screw that holds the axle of the filter wheel. Turn the
filter wheel into a position until there is no filter coding flag in the light barrier.
– Pull off the filter wheel with the axle.
 Important
The lights at the workstation must be dimmed so that the multiplier is not overloaded.
Check the temperature indicator on the service packaging or the multiplier.
If the indicator reads over 54°C, the new multiplier should not be used.
– Pull off the silver protective foil from the new multiplier module and check the convex lenses for dirt. Clean with a lens
cloth as necessary.

– Mount filter wheel. Turn the dark filter in front of the multiplier.
– Unscrew the two T20 Torx screws that hold the flange of the filter wheel motor and screw it onto the new module in
such a way that the gear wheel of the filter wheel motor catches at a right angle in the teeth of the filter wheel without
play. Now tighten the two screws again.
– In the last step the ground connection is transferred.
– Insert the new multiplier module in such a way that the angled rubber seal is subjected to only light pressure.
Make sure that no cables of the light barrier or the motor are caught beneath the multiplier module and that the
module sits correctly on the base plate with the two adjustment pins.
– Tighten the two T20 Torx screws that hold the multiplier module.
– Connect the two X7 and X8 plugs of the ribbon cable for the multiplier to the PCB Control and position in a bow
shape as shown in the picture below.

– Position the black heat shrink tube in such a way that it functions as an upper and lower edge protection (see picture
11).
– Also connect plug X 207 for the PCB Amplifier to the PCB Control. Also position the cable in a bow shape.
– Position the ground plate correctly and tighten the 4 x T20 Torx screws.
– Adjustment of the realplex module is then carried out with the help of the commissioning software.
See Ch. 3 "Commissioning Software".

– The light barrier is fixed in place with two T10 Torx screws.

Disassembly of filter rotor

To replace the motor for the filter wheel the filter rotor first has to be removed.
– Release the M2 socket-head screw fixing the filter rotor shaft in place.
– Turn the filter rotor to a position where no filter coding vane is in the light barrier.
– Remove the filter rotor.
– The realplex module is then adjusted using the commissioning software. See section 3 "Commissioning software".

Pretesting of Mastercycler ep

Before conversion of the Mastercycler ep to the realplex system the service function "Index Units" is used to determine
the hardware status of the cycler.
The PCB Power unit (Power) must have the index –09 or greater to function with the realplex system.
If the index is smaller than –09, the PCB must be replaced.
If the Thermoblock is not clean after cleaning and testing using Commissioning software , the PCB Launch has an index
smaller than –08 and the Thermoblock an index smaller than –10, the PCB Launch and Thermoblock must be replaced.
If the PCB Launch has an index –08 or greater and the Thermoblock an index –10 or greater, the Thermoblock is only
replaced as required.
It is not possible to interrogate the hardware status of the switching power supply unit using the service function "Index
Units".
If the index of the Power supply unit is smaller than –04 siehe sticker, the Power supply unit has to be modified. The
repair kit 5341 896.004 is used for this purpose.

Upgrading of Mastercycler ep to a Mastercycler ep realplex system

Remove the plug for the panel on the rear of the device and put the panel to one side.
The panel is no longer required.

Disassembly of Heated lid

– Put the heated lid into the rear position and move up.
– Remove the silver cover plate in the Heated lid.
– Remove the strain relief.
– Remove the Rollflex cable from the PCB Heated lid.
– Refit the strain relief and the cover plate.
– Dismantle the rear panel of the device.
– Move down the Heated lid and pull out from the back of the device, making sure that the sprung guide pins do not
get lost.
The heated lid is no longer required.

Disassembly of stop eccenter

– Unscrew the Torx screw to remove the stop.


– Attach a blank label from the mounting set over the taphole of the stop.

– Remove the four stoppers in the top of the realplex module.


– Use the special Torx screwdriver to slightly loosen the four screws of the guide sockets. When doing so, the lever
has to be held in different positions to access the four screws.
– Open and close the lever several times so that the guide sockets properly engage in the centering pins and the
heating foil frame in the adjusting plate.
– Move lever in forward position.
– Then tighten the screws in forward position.
– Move lever in backward position and tighten the screws in backward position securely.
The mechanical retrofitting is thereby complete.

Adjustment of the realplex module is then carried out with the help of the commissioning software (CSW). See Ch. 3
"Commissioning Software".

The outer surfaces of the Mastercycler ep realplex system should be cleaned regularly with a cleaning agent.
If hazardous material contaminates the device, the owner is responsible for appropriate cleaning and decontamination.

9 Maintenance
Returning devices

When returning the Mastercycler ep realplex system, ensure that the system is fully decontaminated and does not pre-
sent any health risk to our service staff.
For further information and a decontamination certificate, please visit www.eppendorf.com. Do also consult your labora-
tory safety officer about a suitable decontamination method.
Please fill in the decontamination form and enclose it with the device if it is to be returned to Eppendorf.

Fluorescing substances falsify measurement result of the Mastercycler ep realplex.

Mastercycler modules

The Mastercycler ep realplex system comprises two modules.


Thermomodul
The lower module is the thermomodule (Mastercycler).
The thermoblock is located inside this module.
realplex module
The upper module is the realplex module.
The frame with heating foil and the highly sensitive optics, etc., are located in this module.
Mastercycler ep realplex system
The Mastercycler ep realplex system may only be shipped in the original Eppendorf packaging.

Disinfection procedure

Disinfection of the Realplex module


The Realplex module is disinfected on the surface only.
If the spray disinfection method is used, make sure the spray mist does not spread out beneath the realplex module, and
thence into the heating frame and highly sensitive optics. This will damage the device and so generate high subsequent
costs.

Disinfection of the thermomodule

When disinfecting the surface of the thermomodule, make sure no liquid enters the blue plug on the
Rollflex cable.

1 If the thermoblock is covered by a PCR plate 96, the plate must be removed.
2 Now the visible surface can be disinfected.
3 Replace the PCR plate 96 in the thermoblock after the disinfection.

Make an entry on the Disinfection Certificate to the effect that the heating foil was not disinfected.

Description: Maintenance of PCR Systems

M 01 Cleaning the device according to instruction manual


 Clean PCR systems as described in the respective operating manual and supplement sheets.

M 02 Analyze device data and write down/delete if necessary


(Note: The individual parameters vary, or are not available, based on the PCR system.)

9 Maintenance
 Analyze the following parameters with the service program:
– Error statistics
– Number of cycle counts
– Number of pulse counts
– Number of program counts

M 03 Keyboard
 Check the keyboard by pressing all keys.

M 04 Visual inspection of display


 Check the display with the corresponding service program ( if available).
 When doing so, pay special attention to the following points:
– Defective pixels
– Background lighting

M 05 Stability
 Check the stability of the device feet.
 They must be replaced if fissures or damages are present.

M 06 Cleaning the internal assemblies


 Disassemble/assemble the PCR systems as described in the respective service manual and
supplement sheets.

M 07 Housing
 Remove dust and soiling with a cleaning brush, or, if necessary, compressed air.
 Pay attention to damages that compromise safety.
 With existing fissures, there is the possibility of unintentional infiltration of liquids. If this occurs, the
housing must be replaced.

M 08 Printed circuit board / current supply:


 Remove dust and soiling with a cleaning brush, or, if necessary, compressed air.

M 09 Heated lid
 Clean dust and soiling with a cleaning brush, or, if necessary, compressed air.
 Clean the slide rails for the heated lid with a lint-free cloth.

M 10 Heated lid for MC pro


 Inspect the flexible pressure element of the vapo protect heated lid for damages.

M 11 Thermoblock
 Remove dust and soiling with a cleaning brush, or, if necessary, compressed air.
 When cleaning a realplex thermoblock, the cleaning procedure described in the instruction manual must
be observed.

M 12 Motor/device fan:
 Remove dust and soiling with a cleaning brush, or, if necessary, compressed air.
 Pay attention to operating noise.
 With the service program, select various voltages for testing. ( if available )

M 13 realplex module
 Clean the frame with heating foil, insulating plate, and lower lens array as described in the respective
service manual.

Electrical safety test

Effective standard
IEC 61010-1 (complies with EN 61010-1)

Measuring device

9 Maintenance
The specifications of the above-mentioned standard must be able to be measured using the measuring device.
 Consult the manufacturer of your measuring device.
Only a properly instructed and duly qualified electrician may carry out electrical safety tests.

Notes concerning the measuring procedure


Measurement is carried out in accordance with EN 61010-1.
Measurement pursuant to any other, country-specific standard (e.g. VDE 701/702) will be allowed only if this standard
specifies the same measuring procedures and limiting values and is subordinated to EN 61010-1.
If the measurement is carried out in accordance with any other standard, attention must be paid to the fact that the
measuring arrangement meets the requirements of EN 61010-1.
In the US, the PE conductor current is measured in accordance with NFPA 99.
Handling of the different measuring devices may vary.
 Consult the operating manual of the measuring device for information about the measuring procedure.
 Measurement is carried out on a grounded, metallic point that is not insulated and can be touched from the outside
(e.g. housing screws).
 Never use an optionally provided interface for measurement. You might damage the electronic system of the device.
