RH GROUPING
Routine Rh grouping for donor and patients involve detection of only the D antigen on
red cells. Blood samples are Rh (D) positive or Rh (D) negative depending on the
absence of the D antigen, respectively. However, red cell i as C, E, c, e belong to this
second most important blood group system: Rh system. Their detection is equally
important among individuals because if they are exposed to Rh antigens which they lack,
this results to antibody production and subsequent transfusion of blood having the Rh
antigens will bring about transfusion reaction. The five major Rh antigens in decreasing
immunogenicity are as follows: D,c,E, C,e (Harmening, 2005).
Materials Needed
Glass slides
Marking pen
Lancet (with 3 mm blade)
70% alcohol
Gum label
Applicator sticks
Cotton
Rh view box (surface temperature: 45-50°C)
Reagents/Sample
Anti-D serum
Anti-C serum
Anti-c serum
Unknown blood sample

Rh/Hr Control (CTRL)
Procedure
1. Prepare 2 glass slides
3. Label the 3 portions of the first slide as D, C, and c. Label the 3 portions of the as E, e, and Ctrl.
4. Place the glass slides on Rh view box (surface temperature 45-50C) for 5 minutes
5. Remove the pre-warmed slides and deliver 1 drop of anti-D, anti-C, Anti-E, anti-e, and Rh/Hr control on the appropriately marked portion of the
glass slide.
6. Perform skin puncture by prick method and add 1 drop of whole blood to each portion

Observation
Draw and color your results. Label the slides completely then interpret me determine the
Rh group.
