Test-Retest Reliability and Validity of TecnoBody D-Wall to Assess the Range of Motion During Overhead Squat in Healthy Individuals

This study evaluated the validity and reliability of the TecnoBody D-Wall system in assessing joint range of motion (ROM) during overhead squat movements in healthy individuals, using Kinovea as a reference tool for data comparison. A total of 29 participants (16 males, 13 females) with a mean age of 28.41 ± 6.66 years were included. Measurements were conducted for hip and knee joint angles in the sagittal plane, with three repetitions per participant analyzed using both systems. The D-Wall system employed a 3D Kinect V2 camera and force platform, while Kinovea used 2D video-based motion analysis. The results demonstrated excellent agreement between the two systems, with intra-class correlation coefficients (ICC) ranging from 0.79 to 0.99. For the knee joint, the test-retest ICC values were 0.99 (95% CI: 0.97–0.99) for Kinovea and 0.98 (95% CI: 0.95–0.99) for the D-Wall on the right side, and 0.98 (95% CI: 0.97–0.99) for Kinovea and 0.88 (95% CI: 0.79–0.94) for the D-Wall on the left side. For the hip joint, test-retest ICC values were 0.99 (95% CI: 0.97–0.99) for Kinovea and 0.94 (95% CI: 0.88–0.97) for the D-Wall on the right side, and 0.98 (95% CI: 0.97–0.99) for Kinovea and 0.93 (95% CI: 0.87–0.97) for the D-Wall on the left side. Bland–Altman plots confirmed good agreement, with no significant systematic bias observed. Both systems showed statistically insignificant differences (p > 0.05) between measurements, and correlation values ranged from 0.83 to 0.99, indicating strong associations. These findings highlight the high validity and reliability of both TecnoBody D-Wall and Kinovea systems in measuring joint angles during dynamic movements. The comparable accuracy between systems suggests that either system can be effectively utilized in clinical or research settings, depending on specific needs and resource availability. Full article

Studying exoplanet atmospheres is essential for assessing their potential to host liquid water and their capacity to support life (their habitability). Each atmosphere uniquely influences the likelihood of surface liquid water, defining the habitable zone (HZ)—the region around a star where liquid water can exist. However, being within the HZ does not guarantee habitability, as life requires more than just liquid water. In this study, we adopted a two-pronged approach. First, we estimated the surface conditions of planets near the HZ’s inner edge under various atmospheric compositions. By utilizing a 3D climate model, we refined the inner boundaries of the HZ for planets with atmospheres dominated by H₂ and CO₂ for the first time. Second, we investigated microbial survival in these environments, conducting laboratory experiments on the growth and survival of E. coli K-12, focusing on the impact of different gas compositions. This innovative combination of climate modeling and biological experiments bridges theoretical climate predictions with biological outcomes. Our findings indicate that atmospheric composition significantly affects bacterial growth patterns, highlighting the importance of considering diverse atmospheres in evaluating exoplanet habitability and advancing the search for life beyond Earth. Sepsis is a clinical condition causing tissue damage as a result of infection and an exaggerated immune response. Sepsis causes 11 million deaths annually, a third of which are associated with acute lung injury (ALI). Rapid and effective treatment is crucial to improve survival rates. Punica granatum (pomegranate) is rich in polyphenols and demonstrates strong antioxidant activity, while amifostine acts as a free radical scavenger. This study aimed to investigate the antioxidant and anti-inflammatory effects of P. granatum peel extract (PGPE) and amifostine in sepsis-related ALI. Experimental groups included Control, CLP (cecal ligation and puncture-induced sepsis), Amf (200 mg/kg amifostine, intraperitoneally), and PGPE250, and PGPE500 (250 and 500 mg/kg PGPE via oral gavage, respectively). Thiobarbituric acid reactive substances (TBARS), total thiol (TT), tumor necrosis factor-alpha (TNF-α) levels, and metalloproteinases 2 and 9 (MMP-2 and MMP-9) were assessed in the lung tissue. Biochemical analysis demonstrated that TBARS and TNF-α levels significantly decreased in both the PGPE and amifostine treatment groups compared to the CLP group, while TT levels showed notable improvement. Histopathological evaluation revealed reduced MMP-2 and MMP-9 immunopositivity in the PGPE250 and PGPE500 groups. These findings highlight the lung-protective properties of PGPE, underscoring its potential as a therapeutic agent for sepsis-induced acute lung injury. Full article

Aortic arch anomalies represent a range of congenital vascular malformations resulting from disruptions in the typical embryological development of the aortic arch and its branches. These anomalies, which vary widely in their presentation, can lead to significant clinical symptoms depending on their structure and position. We report the case of a 75-year-old male with intermittent hypertension, palpitations, and episodic warmth in the upper body. Computed tomography (CT) angiography revealed an atypical aortic arch anatomy with a unique right subclavian artery anomaly. The aortic arch displayed a typical orientation but included an additional arterial branch arising from the medial wall of the descending aorta. This aberrant branch with a tortuous aspect coursed posteriorly around the esophagus and merged with the subclavian branch of the brachiocephalic trunk, forming a vascular ring. A possible embryological hypothesis requires the persistence of both the distal segment of the right dorsal aorta and the right seventh intersegmental artery, as well as the right fourth aortic arch; however, the imaging aspect of our patient is not that of a classic double aortic arch. This case emphasizes the importance of advanced imaging techniques, such as CT angiography, in identifying and managing rare vascular anomalies that may influence patient care and clinical outcomes. Therapy for Dupuytren’s Disease (II): Collagenase Therapy vs. Limited Fasciectomy—A Long-Term Comparative Study

Background: Dupuytren’s disease (DD) is a systemic connective tissue disorder of the palm, predominantly affecting men of Northern European or Caucasian origin over 55. In addition to conventional surgery, Dupuytren’s contracture can be treated in a minimally invasive way by injecting bacterial collagenase into the cord. However, studies on the long-term success rate when compared to the gold standard, surgical limited fasciectomy, are limited. Methods: This monocentric retrospective study examined 35 patients who had been treated with bacterial collagenase for Dupuytren’s contracture, conducting a long-term follow-up after an average of 5.7 years. The results were compared to a control group of 40 patients treated with surgical limited fasciectomy on average 5.5 years ago. Finger extension (Tubiana stage), strength, sensitivity, the effect of possible risk factors, and patient-reported outcome measures (PROMs) were compared between the two groups. Results: The long-term results after therapy for DD showed a significant reduction in the Tubiana stage for both groups (p < 0.001). Additionally, we observed a longer mean preintervention Tubiana stage and a better long-term improvement in the Tubiana stage for patients with limited fasciectomy when compared to the collagenase group. (both p < 0.001). Neither grip strength nor the pinch test showed significant differences when compared within each group or when comparing both groups. Both the treated and untreated fingers of patients with limited fasciectomy had a superior two-point discrimination (p < 0.001). For the URAM questionnaire, we observed a significantly better result in the control group (p < 0.01). Retrospectively, significantly more patients in the collagenase group would not choose the same therapy to treat DD (35 vs. 8%; p < 0.05). Conclusions: The two therapy options should be seen as complementary for the treatment of DD. Collagenase therapy seems a sensible option for DD with an earlier Tubiana stage and contractures that predominantly affect the MCP joint. Contractures with higher Tubiana stages that also affect the PIP joint should predominantly be treated with limited fasciectomy. Full article

Carl Woese’s thesis of cellular evolution emphasized that the last universal common/cellular ancestor (LUCA) must have evolved by drawing from “global inventions”. Yet, existing theories regarding the origin(s) of LUCA have mostly centered upon scenarios that LUCA had evolved mostly independently. In an earlier paper, we advanced a new theory regarding the origin(s) of LUCA that extends Woese’s original insights. Our theory centers upon the possibility that different vesicles and protocells can merge with and acquire each other as a form of variation, selection, and retention, driven by wet-and-dry cycles and other similar cyclical processes. In this paper, we use computer simulation to show that under a variety of simulated conditions, LUCA can indeed be produced by our proposed processes. We hope that our study can stimulate laboratory testing of some key hypotheses that vesicles’ absorption, acquisition, and merger has indeed been a central force in driving the evolution of LUCA. In this study, the impact of leonardite as a feed additive in goldfish diets was examined, focusing on its potential to positively influence feed and water parameters, which are critical for achieving sustainable ornamental fish farming. In this study, goldfish were fed diets containing four different levels of leonardite (0%, 2%, 6%, 10%). The experiment was conducted in 12 tanks with 3 replicates per treatment and lasted for 3 months. The fish, which were provided from the Research, Production and Training Institute, with an average live weight of 0.947 ± 0.03 g and an average length of 1.12 ± 0.02 cm, were used. After 90 days, it was observed that the specific growth rates increased in correlation with the amount of leonardite in the diet, with a statistically significant difference identified (p < 0.05). In this study, which investigated the coloration of fish with leonardite added to the feed for the first time, it was determined that the color of the fish approached from yellow to orange as the amount of leonardite added to the feed increased (p < 0.05). Given the positive effects observed on pigmentation and growth in this study, it is recommended that further detailed investigations be conducted on leonardite, a natural, harmless, and cost-effective additive, using different fish species. Blood pressure measurement is important in monitoring hypertension. However, blood pressure does not provide much information about renal condition in treated hypertension. This study aimed to evaluate renal oxygenation in hypertensive patients using T2* mapping. Subgroup analysis explored whether R2* values can guide adjustments in antihypertensive treatment. A total of 140 consecutive subjects were recruited: 87 hypertensive subjects and 53 normotensive subjects. Hypertensive subjects were classified into non-medication (non-med), angiotensin II receptor blocker (ARB), and non-ARB-treated groups. Each group was divided into good and poor control subgroups based on blood pressure at enrollment. T2* mapping was utilized to assess renal cortical and medullary R2* values. After a 2-year follow-up, subjects were categorized into stable and unstable based on the need for treatment modifications. The unstable subgroup had higher medullary R2* values than the stable subgroup in all followed patients (p < 0.05). Additionally, the unstable merged non-med with ARB subgroup had higher medullary R2* values overall (p < 0.05) and within the good control subgroup (p < 0.05). Patients with stable hypertension, especially those with good control managed through lifestyle modifications or ARBs, exhibited lower renal medullary R2* values, suggesting higher renal oxygenation. Background: In the context of modern cancer therapy, the management of adverse effects of systemic therapies can lead to the avoidance of underdosing and withdrawal and increases in the quality of the therapeutic act and the quality of life. This review offers an overview of the skin-related toxicities associated with Cabozantinib, a multikinase inhibitor (MKI) that is approved for treating advanced kidney cancer, hepatocellular carcinoma, and medullary thyroid cancer. It covers the most common dermatological side effects, such as palmar–plantar erythrodysesthesia, stomatitis, hair alterations, xerosis, scrotal erythema, and subungual splinter hemorrhages. Additionally, this review includes suggested preventive strategies and management approaches based on the severity of these adverse effects. Full article

Background: The ketogenic diet (KD), high in fat and low in carbohydrates, was introduced in the 1920s as a non-pharmacological treatment for refractory epilepsy. Although its mechanism of action is not fully understood, beneficial effects have been observed in neurological diseases such as epilepsy, Alzheimer’s disease, and Parkinson’s disease. Objective: This review examines the impact of the ketogenic diet and its molecular and neuroglial effects as a complementary therapy for neurological diseases. Discussion: KD is associated with neuroprotective and antioxidant effects that improve mitochondrial function, regulate neurotransmitter flow, and reduce neuroinflammation and oxidative stress. Glial cells play an essential role in the utilization of ketone bodies (KBs) within the central nervous system’s metabolism, particularly during ketosis induced by the KD. Thus, the KD represents a broad and promising strategy that involves both neurons and glial cells, with a molecular impact on brain metabolism and neuroinflammatory homeostasis. Conclusion: Multiple molecular mechanisms have been identified to explain the benefits of the KD in neurological diseases; however, further experimental and clinical studies are needed to address various molecular pathways in order to achieve conclusive results. Extracellular vesicles (EVs) are nanosized, membrane-bound structures that have emerged as promising tools for drug delivery, especially in the treatment of lysosomal storage disorders (LSDs) with central nervous system (CNS) involvement. This review highlights the unique properties of EVs, such as their biocompatibility, capacity to cross the blood–brain barrier (BBB), and potential for therapeutic cargo loading, including that of enzymes and genetic material. Current therapies for LSDs, like enzyme replacement therapy (ERT), often fail to address neurological symptoms due to their inability to cross the BBB. EVs offer a viable alternative, allowing for targeted delivery to the CNS and improving therapeutic outcomes. We discuss recent advancements in the engineering and modification of EVs to enhance targeting, circulation time and cargo stability, and provide a detailed overview of their application in LSDs, such as Gaucher and Fabry diseases, and Sanfilippo syndrome. Despite their potential, challenges remain in scaling production, ensuring isolation purity, and meeting regulatory requirements. Future developments will focus on overcoming these barriers, paving the way for the clinical translation of EV-based therapies in LSDs and other CNS disorders. Full article

Regulation of LPS-Induced Inflammatory Responses in Bovine Mammary Epithelial Cells via TLR4-Mediated NF-κB and MAPK Signaling Pathways by Lactoferrin

Lactoferrin (LF), a member of the transferrin family, is widely present in mammalian milk and other secretions, exhibiting anti-inflammatory, antibacterial, and anti-infective properties. Although the biological functions of LF have been extensively studied, there are few reports on its effects and molecular mechanisms concerning bovine mastitis caused by bacterial infection. This study used bovine mammary epithelial cells (BMECs) cultured in vitro as the research model. An inflammatory injury model was established by stimulating BMECs with LPS to investigate whether LF at different concentrations (10, 50, 100, and 200 μg·mL⁻¹) could inhibit the inflammatory response before and after the onset of inflammation. The expression of inflammatory cytokines IL-1β, IL-6, IL-8, and TNF-α at both the gene and protein levels was detected using RT-qPCR and ELISA. Western blotting was employed to evaluate the phosphorylation levels in the inflammatory signaling pathways MAPK/P38/ERK and NF-κB/P65, while RT-qPCR was used to examine the impact on TLR4 receptor gene expression. The results display that pretreatment with LF prior to LPS-induced inflammation in BMECs reduced the expression of inflammatory cytokines IL-1β, IL-6, IL-8, and TNF-α at both the gene and protein levels (p < 0.05). LF also inhibited the phosphorylation of NF-κB/P65 and MAPK/P38/ERK signaling pathways and downregulated TLR4 receptor gene expression (p < 0.05). However, when LF was added after the onset of LPS-induced inflammation, inflammatory cytokine expression and phosphorylation levels in the NF-κB/P65 and MAPK/P38/ERK pathways remained elevated, along with high expression of the TLR4 receptor gene (p < 0.05). These findings show that LF can antagonize LPS-induced inflammatory responses in BMECs and reduce cytokine expression, exhibiting anti-inflammatory effects when administered before inflammation. Conversely, when LF is added post-inflammation, it appears to enhance cytokine expression, potentially promoting the recruitment of more cells or factors to resolve inflammation rapidly. Both effects are mediated through the TLR4 receptor and the NF-κB/P65 and MAPK/P38/ERK signaling pathways. Full article

Background: several authors have documented variations in local temperature in both horses and dogs presenting acute intervertebral disc extrusion (IVDE) along the entire spinal column. However, none have demonstrated distinct temperature differences between healthy animals and those with IVDE. A retrospective study was conducted to assess the efficacy of thermography at evaluating local temperature and thermal patterns in healthy dogs as well in those with IVDE across the T11–L3 area. Methods: the study included 20 healthy dogs and 32 dogs with IVDE. For both groups of dogs, the thoracolumbar region was trimmed and, subsequently, scanned using the Flir E50 thermography device. The Flir Tool software was used to analyze three designated areas (Bx1, Bx2, Bx3) within the thoracolumbar region by comparing the average temperature of the minimum, maximum, and mean temperature recordings between the two groups. Results: the thermal pattern and the local temperature of the thoracolumbar area present differences between healthy dogs and those with IVDE. Conclusions: we recommend thermographic scanning of the thoracolumbar area to find differences in local temperature between healthy dogs and those with intervertebral disc extrusion. Further investigations are required to differentiate between disc extrusion that exhibits lateralization to the right or left. Full article

Capsella bursa-pastoris Medik. (CBP) is a species with antibacterial, anti-inflammatory, antioxidant, anticancer, and hepatoprotective effects. We have chosen to study this species because, although it is a common plant with a distinctive fruit appearance, its effects are not fully understood. The aim of this study was to characterize the histoanatomy of the vegetative, reproductive organs and to characterize CBP extracts in terms of bioactive compounds and its antioxidant capacity. This study investigated the quantitative chemical composition of this species using the HPLC method, revealing the total content in polyphenols, flavonoids, and anthocyanins, and investigated the antioxidant potential through fluorescence recovery after photobleaching (FRAP assay), cupric ion (Cu²⁺) reduction, (CUPRAC assay), and a free radical scavenging method (DPPH). Our results show that CBP is a rich source of flavonoids, mainly from the extract obtained from the fruits; it has an antioxidant capacity, with the highest values being obtained from mature flowers and ripe fruits. Of the active principles, the highest amounts, according to HPLC determinations, were obtained in flowers and are represented by hyperoside. Thus, we can recommend the studied species for phytopharmaceutical preparations. Full article

Pilot Study on AI Image Analysis for Lower-Limb Reconstruction—Assessing ChatGPT-4’s Recommendations in Comparison to Board-Certified Plastic Surgeons and Resident Physicians

AI, especially ChatGPT, is impacting healthcare through applications in research, patient communication, and training. To our knowledge, this is the first study to examine ChatGPT-4’s ability to analyze images of lower leg defects and assesses its understanding of complex case reports in comparison to the performance of board-certified surgeons and residents. We conducted a cross-sectional survey in Switzerland, Germany, and Austria, where 52 participants reviewed images depicting lower leg defects within fictitious patient profiles and selected the optimal reconstruction techniques. The questionnaire included cases with varied difficulty, and answer options did not always include the most obvious choices. Findings highlight that ChatGPT-4 successfully evaluated various reconstruction methods but struggled to determine the optimal solution based on the available information in visual and written forms. A chi-squared test of independence was performed to investigate the overall association between answer options (A, B, C, and D) and rater group (board-certified surgeons, ChatGPT-4, and resident). Inter-group rater associations showed significant overall test results (p < 0.001), with high agreement among board-certified surgeons. Our results suggest that board-certified plastic surgeons remain essential for patient-specific treatment planning, while AI can support decision-making. This reaffirms the role of AI as a supportive tool, rather than a replacement, in reconstructive surgery. Full article

Background: The aim of this study was to evaluate visual outcomes and patient satisfaction after bilateral implantation of a new hydrophobic acrylic intraocular lens called Clareon (Alcon) using the mini-monovision technique. Methods: A single-center, prospective, nonrandomized study was conducted in Tandil (Buenos Aires, Argentina), including patients scheduled for cataract surgery. To achieve mini-monovision, the spherical equivalent was calculated between −0.25 and +0.25 D for the dominant eye, and between −0.75 and −1.00 D for the non-dominant eye. The main outcomes were uncorrected distance visual acuity (UDVA) and uncorrected intermediate visual acuity (UIVA) evaluated at 66 cm. A secondary outcome, patient satisfaction, was assessed using the CatQuest-9SF questionnaire. Results: The mean binocular UDVA was 0.01 ± 0.05 logMAR three months after surgery, while the mean binocular UIVA was 0.20 ± 0.06 logMAR. The postoperative mean spherical equivalent in the dominant eye was −0.27 ± 0.12, and in the non-dominant eye was −0.87 ± 0.25. Before surgery, the CatQuest-9SF questionnaire revealed that 83.33% of patients were dissatisfied with their vision during daily activities. Over 50% reported significant difficulties with reading newspapers, sewing, and reading TV subtitles. Additionally, 66.6% struggled with recognizing faces, 50% with seeing product prices, and 50% with walking on uneven ground. Post-surgery, most patients experienced improved vision for daily tasks, with no reports of high dissatisfaction or significant difficulties. Patients were quite satisfied with their vision for hobbies and TV subtitles, and very satisfied (90%) with seeing supermarket prices. Conclusions: patients implanted with a new enhanced monofocal IOL using the mini-monovision technique showed improved distance and intermediate visual acuity, reduced need for glasses, and expressed a high degree of satisfaction. High and Low Adherence to Mediterranean and DASH Diet Patterns and the Risk of Heart Failure: A Meta-Analysis of Observational Studies

Background. The relationship between heart failure (HF) and Mediterranean and DASH diets is not well delineated. This meta-analysis aimed to assess the effectiveness of high adherence to Mediterranean and DASH diets compared to low adherence in reducing the risk of incident HF (primary prevention of HF) and reducing all-cause mortality in patients with HF (secondary prevention of HF). Methods. The reporting stages of this meta-analysis closely adhered to the PRISMA guidelines. A comprehensive literature search was undertaken for published papers in PubMed, Embase, EBSCO, ICTRP, and the NIH clinical trials databases. Results. A total of 16 reports from 14 studies were included in this paper. A significant inverse association was identified between high adherence to the Mediterranean diet model (compared to low adherence) and the risk of incident HF (OR = 0.77, 95% CI: 0.63–0.93, p = 0.007) among patients without previous diagnosis of HF. Similarly, there was a significant and inverse relationship between high adherence to the DASH diet (compared to low adherence) and the risk of incident HF (OR = 0.83, 95% CI: 0.70–0.98, p = 0.03) among patients without previous diagnosis of HF. High adherence to the Mediterranean diet model (compared to low adherence) was associated with lower all-cause mortality (OR = 0.88, 95% CI: 0.78–0.99, p = 0.03) among patients with HF. Conclusions. This paper demonstrated that high adherence to Mediterranean and DASH diets significantly reduced the risk of incident HF among individuals without a previous diagnosis of HF, whereas only high adherence to the Mediterranean diet was associated with lower all-cause mortality among patients with HF. Estimation Model for Cotton Canopy Structure Parameters Based on Spectral Vegetation Index

The spectral vegetation indices derived from remote sensing data provide a detailed spectral analysis for assessing vegetation characteristics. This study investigated the relationship between cotton yield and canopy spectral indices to develop yield estimation models. Spectral reflectance data were collected at various growth stages using an ASD FieldSpec Pro VNIR 2500 spectrometer. Six prediction models were developed using spectral vegetation indices, including the Normalized Difference Vegetation Index (NDVI) and Ratio Vegetation Index (RVI), to estimate the Leaf Area Index (LAI) and above-ground biomass. For LAI estimation using the NDVI, the power function model (y = 10.083x^11.298) demonstrated higher precision, with a multiple correlation coefficient of R² = 0.8184 and the smallest root mean square error (RMSE = 0.3613). These results confirm the strong predictive capacity of NDVI for LAI, with the power function model offering the best estimation accuracy. In estimating above-ground biomass using RVI, the power function model of y = 6.5218x^1.33917 achieved the higher correlation (R² = 0.8851) for fresh biomass with an RMSE of 0.1033, making it the most accurate. For dry biomass, the exponential function model (y = 9.1565 × 10⁻⁵∙exp(1.1146x)) was the most precise, achieving an R² value of 0.8456 and the lowest RMSE value of 0.0076. These findings highlight the potential of spectral remote sensing for accurately predicting cotton canopy structural parameters and biomass weights. By integrating spectral analysis techniques with remote sensing, this research offers valuable insights for precision cotton planting and field management, enabling optimized agricultural practices and enhanced vegetation health monitoring.
