Instruction On Using Standard Particle
This instruction will introduce the different standard particles, different version of
software and optical system. And then tell you how to use the standard particle for
different version of software and optical system.
Standard particles
Standard particles could be used to check and adjust the optical system. There are
particles of different diameters used on BC-5500.
We recommend two brands of 10um standard particle to you, Duke and Beckman Coulter.

Software version:
Check the software version you are using.
Click the “Status” button at the bottom of the “Service” screen to enter the Version
screen shown in the figure below.
If the version of “App software” is lower than 1.5.0.1696, the coordinates of
scattergram in the “OpAdjust” screen is 250.
Brief summary:
For the old optical system, 10um standard particle is used to check the DIFF channel;
7um standard particle is used to check the BASO channel. There are different target
values of standard particle test for different software version.
For the new optical system, 7um standard particle is used for both channels (DIFF &
BASO).
2) In the “OpAdjust” screen, click “calculate”, and then check the four parameters of
calculation results (the default smooth histogram is 5-Point Mean). Check the DIFF
channel whether it meets the 5 requirements (quantitative): MAS peak: 154 ± 4; LAS
peak: 79 ± 4; MAS 0.1max width: 12-20; LAS 0.1max width: 8-12; Width of MAS
should be greater than that of LAS. Please note that for the “App software” version:
1.5.0.1696 or more, the coordinates in the “OpAdjust” screen have been reduced to
176 and some of the requirements are different: MAS Peak: 106 ± 4; LAS peak: 54 ±
4.
3) If the MAS/LAS peak value is out of normal range, input the target value (depends on the
software version; Parti1 LAS: 79 or 54, Parti1 MAS: 154 or 106) and press “>>>” to
calculate the new Gain. Please write down the integer part of new Gain; then access
“Setup > Gain” screen to modify the WBC_LAS(DIFF) and WBC_MAS(DIFF) Gain
accordingly; set them as the integer part of new Gain.
6) Select the “BASO” channel and click "Calculate" to check the (only) two parameters:
Mas Peak, 147 ± 4; LAS Peak, 58 ± 4.
7) If the MAS/LAS peak value is out of normal range, input the target value and press
“>>>” to calculate the new Gain. Please write down the integer part of new Gain; then
access “Setup > Gain” screen to modify the Gain accordingly; set them as the integer part
of new Gain.
