Instruction on using the standard particle

This instruction will introduce the different standard particles, different version of
software and optical system. And then tell you how to use the standard particle for
different version of software and optical system.

Standard particles
Standard particles could be used to check and adjust the optical system. There are
particles of different diameters used on BC-5500.

10um standard particle:

We recommend two brands of 10um standard particle to you, Duke and Beckman Coulter.

7um standard particle:

7um standard particle from Duke is recommended.

Software version and Optical system

Software version:
Check the software version you are using.
Click the “Status” button at the bottom of the “Service” screen to enter the Version
screen shown in the figure below.
If the version of “App software” is lower than 1.5.0.1696, the coordinates of
scattergram in the “OpAdjust” screen is 250.

If the version of “App software” is 1.5.0.1696 or more, the coordinates of scattergram


in the “OpAdjust” screen is reduced to 176.
Optical system:
Identify if you are using the new version of optical system or not.
The machines with the serial number before Rx75-0360 are with old optical systems.
New optical system is equipped with constant temperature system such as heater etc.
The picture below shows you the new optical system with constant temperature
system.

Brief summary:

For the old optical system, 10um standard particle is used to check the DIFF channel;
7um standard particle is used to check the BASO channel. There are different target
values of standard particle test for different software version.

For the new optical system, 7um standard particle is used for both channels (DIFF &
BASO).

How to use standard particle

Old optical system


10um standard particle test (DIFF channel):
1) Add several (2 or 3) drops of 10um standard particles into a centrifugal tube. Enter the
“Count” screen, run the 10um standard particles as whole blood sample in the
OV-WB-CBC+5DIFF mode. After that, enter the “Review” screen to select test data of
the 10um standard particles, and click “OpAdjust” (“optical adjustment”) to determine the
standard particles’ shape on the scattergram (qualitative). The shape shall be a level strip
or a strip tilting slightly (the right end tilting shall be within 30 degrees).
The normal scattergrams of 10um standard particles are shown as below.

2) In the “OpAdjust” screen, click “calculate”, and then check the four parameters of
calculation results (the default smooth histogram is 5-Point Mean). Check the DIFF
channel whether it meets the 5 requirements (quantitative): MAS peak: 154 ± 4; LAS
peak: 79 ± 4; MAS 0.1max width: 12-20; LAS 0.1max width: 8-12; Width of MAS
should be greater than that of LAS. Please note that for the “App software” version:
1.5.0.1696 or more, the coordinates in the “OpAdjust” screen have been reduced to
176 and some of the requirements are different: MAS Peak: 106 ± 4; LAS peak: 54 ±
4.
3) If the MAS/LAS peak value is out of normal range, input the target value (depends on the
software version; Parti1 LAS: 79 or 54, Parti1 MAS: 154 or 106) and press “>>>” to
calculate the new Gain. Please write down the integer part of new Gain; then access
“Setup > Gain” screen to modify the WBC_LAS(DIFF) and WBC_MAS(DIFF) Gain
accordingly; set them as the integer part of new Gain.

7um standard particle test (BASO Channel):


1) Select "OV-PD" mode. And click "Diluent" in the “Count” screen to dispense diluent into
a centrifugal tube 3 times. Then exit the diluent dispense screen.
2) Dispense 1 drop of 7um standard particle into the centrifugal tube and mix them.
3) Change the mode to "OV-Whole blood-CBC+5DIFF" and test the dilution as whole
blood.
4) Then go to the "Review > OpAdjust" screen and select the "BASO" channel.
5) Click "Calculate" to check the (only) two parameters: MAS Peak and LAS Peak. The
range is listed as following: Mas Peak: 70 ± 4, LAS Peak: 81 ± 4. Please note that for
the “App software” version: 1.5.0.1696 or more, the coordinates in the “OpAdjust”
screen have been reduced to 176 and the requirements are different: MAS Peak: 43
± 4; LAS peak: 56 ± 4.
6) If the MAS/LAS peak value is out of normal range, input the target value (depends on the
software version; Parti1 LAS: 81 or 56, Parti1 MAS: 70 or 43) and press “>>>” to
calculate the new Gain. Please write down the integer part of new Gain; then access
“Setup > Gain” screen to modify the WBC_LAS(BASO) and WBC_MAS(BASO) Gain
accordingly; set them as the integer part of new Gain.
Actually, for the BASO channel, only need to make sure the lower part of BASO
scattergram should be smooth and not cut by a straight line when testing the normal
fresh sample. If the BASO scattergram is cut, please increase the BASO Gain
manually and run normal fresh sample to confirm. The pictures below show the
normal scattergram of sample.

“App Software” version lower than 1.5.0.1696:

“App Software” version 1.5.0.1696 or more:

New optical system


7um standard particle test (DIFF & BASO channel):
1) Select "OV-PD" mode. And click "Diluent" to dispense diluent into a centrifugal tube 3
times.
2) Dispense 1 drop of 7um standard particle into the centrifugal tube and mix them.
3) Change the mode to "OV-Whole blood-CBC+5DIFF" and test the dilution as whole
blood.
4) Then go to the "Review > OpAdjust" screen and select the "DIFF" channel.
5) Cilck "Calculate" to check the four parameters: Mas Peak, 88 ± 4; LAS Peak, 45 ± 4;
MAS 0.1max width, ≤14; LAS 0.1max width, ≤14.

6) Select the “BASO” channel and click "Calculate" to check the (only) two parameters:
Mas Peak, 147 ± 4; LAS Peak, 58 ± 4.
7) If the MAS/LAS peak value is out of normal range, input the target value and press
“>>>” to calculate the new Gain. Please write down the integer part of new Gain; then
access “Setup > Gain” screen to modify the Gain accordingly; set them as the integer part
of new Gain.
