Starch Test

The document describes steps to test a green leaf for starch. The leaf is boiled in water to soften it, then placed in ethanol in a test tube in hot water for 10 minutes. This removes the chlorophyll from the leaf. The leaf is then placed in iodine solution, which will color the leaf blue-black if starch is present, indicating the plant was photosynthesizing.

Testing a green leaf for starch

Aim
To test a green leaf for starch, which is a sign that the plant has been photosynthesising.

Step 1 Place a leaf in boiling water Step 2 Turn off the Bunsen burner.

for about 1 minute to soften it.  Place the leaf into a test tube
half full of ethanol.
 Place the test tube in the hot water for
10 minutes.

Step 4 Spread the leaf out on a Petri dish

and cover it with iodine solution.

Step 3 Return the leaf to the hot water for

about a minute.

1 What colour did your leaf go when the iodine was added? ...............................................................
2 What result would you expect if the plant had been kept in the dark for 48 hours?

............................................................................................................................................................
3 The ethanol removed the chlorophyll from the leaf. Why was this necessary?

............................................................................................................................................................
4 Why does the ethanol continue to boil, even though the Bunsen burner is turned off?
