Activity 4-1

AMINO ACID SEQUENCE AND

PROTEIN STRUCTURE AND DENATURATION

Most Essential Learning Outcomes

At the end of the activity, students must have:
1. described amino acid and proteins
2. discussed the cellular functions of proteins
3. identified, described and differentiated essential from non-essential amino acids
4. described the structures of proteins

I. Amino Acid Sequence

Directions
The following Data Table 1 shows the amino acids found in selected sites in
hemoglobin of different vertebrates. Examine the chart for differences between humans and
other organisms.
1. Count the numbers of molecules of each amino acid in human hemoglobin. Record
these totals in the appropriate column of Data Table 2.
2. Count the number of molecules of each amino acid of other vertebrates’ hemoglobin.
Record these totals in the appropriate columns of Data Table 2.
3. Going from left to right, note the position of each amino acid. Count the number of
similarities in the amino acid positions in human hemoglobin as compared with the
hemoglobin of the other vertebrates in Data Table 1. Record your observations in Data
Table 3.
4. Reexamine Data Table 1 and count the numbers of differences in the amino acid
positions in human hemoglobin as compared with the hemoglobin of the other
vertebrates in Data Table 1. Record observations in Data Table 3.

Date Table 3.

Questions for discussion:

1. What can you say about the amino acid sequences of different organisms?
2. Which primate is closely related to humans?
3. Which non-primate is closely related to humans?
4. Discuss the importance of amino acid sequence.

II. Protein Structure

State which structure (or structures) best relates to the description: primary 1o, secondary 2o,
tertiary 3 o, quaternary 4o. You may use more than one.

III. Hydrolysis and Denaturation

State whether description related to denaturation or hydrolysis by writing the correct term on
the second column
