The Naval Research Laboratory has developed a magnetic microbead support that enables the capture of genomic targets in a single step. The microbead is functionalized with a phosphorous branched dendrimer linker, which covalently binds DNA capture probes, allowing hybridization of the target directly onto the bead. The targets can then be recovered through heat denaturation in small elution volumes for direct analysis. The robust bond between capture probe and microbead permits the regeneration of the probe even after repeated cycles at high temperatures (up to 100C) and salt conditions (3-5 molar NaCl), providing a reusable universal platform for sequence-capture or background subtraction. Use of variable branched dendrimeric linker systems enables the performance to be tailored for specific targets.
