Aim: Determination of PA2 value of prazosin using rat anococcygeus muscle (by Schilds plot

method).

1. Mount the tissue on tissue organ bath with Tyrode solution

2. After relaxation of tissue, record DRC of noradrenaline

A1-A5 is 0.1, 0.2, 0.4, 0.8, 1.6 ml dose of Noradrenaline (100 ug/ml) respectively,

3. Select one dose norepinephrine which falls between 25-75% in DRC label it as 2A.

4. Record 2A response norepinephrine in presence of increase concentration of antagonist i.e.


prazocin till 2A become A or less than A.

A=0.2ml

2A=0.4ml

B1= 5 ug/ml prazosin after 15 min add 0.4 ml of 2A,then measured response height. After
measuring response without washing add 2A after 5 min time cycle , then again added 2A(0.4 ml) to
get same height it indicate antagonist removed from tissue.

B2=8 ug/ml prazosin after 15 min add 0.4 ml of 2A,then measured response height. After
measuring response without washing add 2A after 5 min time cycle , then again added 2A(0.4 ml) to
get same height it indicate antagonist removed from tissue.
B3=11ug/ml prazosin after 15 min add 0.4 ml of 2A,then measured response height. After
measuring response without washing add 2A after 5 min time cycle , then again added 2A(0.4 ml) to
get same height it indicate antagonist removed from tissue.

So , X=35x5=175

Aim: Determination of PD2 value of Histamine using guinea pig ileum.

Dose Response curve of Histamine (100ug/ml)

1. Make observation table as like DRC


2. Plot log dose response versus % response curve.
3. Determine dose of histamine which produce 50% response .
For example
you get the value from graph at 50% response suppose 1.04
Take antilog(1.04)= 10.96
Concentration 100ug/ml
1 ml=100 ug
X= 10.96
X=10.96/100=0.1096

To determine concentration in of histamine in organ tube which produce 50% response

Y=Quantity in ml to produce 50% response(x) X Concentration of histamine used

Bath capacity (ml)

Y=01096 X 100/32
Y=0.3406 ug/ml

Molar concentration =concentration which produce 50% response /molecular weight X 1000
= 0.3406/111.15X1000
=0.003064/1000
=3.064X10^-6
pD2 Value =log[3.064X10^-6]
=6- log[3.064]
=6-0.4862
=5.5138
