Chemistry Project: Analysis of pH and Protein in Various Shampoos

Introduction

Shampoos are one of the most commonly used personal care products, and their composition can

affect hair health. The two primary factors that contribute to hair and scalp health are the pH value

and the presence of proteins. In this project, we analyze the pH and protein content of five popular

shampoo brands: Dove, Sunsilk, Vatika, TRESemme, and Head & Shoulders.

Aim

The aim of this project is to determine the pH values and detect the presence of proteins in different

shampoo brands and to analyze which shampoos are most suitable for different hair types based on

pH and protein content.

Apparatus

1. pH paper

2. Test tubes

3. Distilled water

4. Biuret reagent for protein detection

5. Concentrated nitric acid (for Xanthoproteic test)

6. Shampoo samples (Dove, Sunsilk, Vatika, TRESemme, Head & Shoulders)

7. Cation and anion testing reagents (for oxalate, acetate, chloride tests)

Theory

The pH of shampoos plays an essential role in maintaining hair health. The ideal pH for shampoos
should be slightly acidic (around 5-6) to match the natural pH of the scalp. Shampoos with higher pH

values can cause hair damage by lifting hair cuticles.

Proteins are another crucial component in shampoos. They help in hair repair by filling in the gaps in

damaged hair shafts. The Biuret test is used for detecting peptide bonds in proteins, where a violet

color indicates the presence of proteins.

Procedure

1. Prepare shampoo solutions by mixing 2 ml of each shampoo with 20 ml of distilled water.

2. Measure the pH of each solution using pH paper.

3. Perform the Biuret test by adding 2 ml of Biuret reagent to each shampoo solution. A violet color

indicates the presence of proteins.

4. Perform the Xanthoproteic test by adding a few drops of concentrated nitric acid and heating in a

water bath. Yellow coloration indicates the presence of proteins.

5. Perform cation-anion tests to detect oxalate, acetate, and chloride ions as part of additional

chemical analysis.

Observations

The following observations were made after performing the tests:

- **Dove**: pH 5, violet color observed in Biuret test, indicating the presence of proteins.

- **Sunsilk**: pH 5, light blue color in Biuret test, weaker protein indication.

- **Vatika**: pH 5, light blue color in Biuret test.

- **TRESemme**: pH 5, light blue color in Biuret test.

- **Head & Shoulders**: pH 5, light blue color in Biuret test.

In the Xanthoproteic test, no yellow color was observed, suggesting the absence of aromatic amino
acids in the shampoos.

Test Tube Observations


Conclusion

Based on the tests performed, it was observed that Dove shampoo had the most intense violet color

in the Biuret test, indicating a higher presence of proteins compared to other brands. The pH of all

shampoos was found to be around 5, which is ideal for hair health. No shampoo tested positive for

aromatic amino acids in the Xanthoproteic test.

Result

The pH of all the shampoos tested was found to be 5. The Biuret test indicated the presence of

proteins in all shampoos, with Dove showing the most intense reaction. No aromatic amino acids

were detected in the Xanthoproteic test.
