LAB E-REPORT
Objectives:
I) To prepare an onion epidermal cell slide.
II) To identify the structure off the onion epidermal cell
Introduction:
Plant tissues are divided into two types; meristematic tissues and permanent
tissues. Permanent tissues consists of mature cells that have specialized
structure and functions. Permanent tissues can be divided into three which
are dermal tissues (epidermal and peridermal), ground tissues (parenchyma,
sclerenchyma, collenchyma and endodermis) and vascular tissues (xylem and
phloem).
Apparatus:
Compound microscope, razor blade/ cutter, slide, cover slip, dropper, petri
dish, dissecting pin, filter paper/tissue paper, forceps, white tiles
Materials:
Onion, 1% iodine solution
(Caution: Be careful while handling razor blade/cutter and chemicals)
Procedures:
1. The slide was cleaned from any dirt.
2. The onion was cut into small pieces.
3. The inner epidermal layer was stripped off from the scale of an onion and
place it on the slide.
4. 1-2 drops of dilute iodine solution was applied on the epidermal layer.
5. The cover slip was lowered carefully by using pin/pen.
6. Tissue/ filter paper was used to absorb excess iodine on the slide.
7. The epidermal tissues was observed using 10x and 40x objective lens
8. The cell wall, nucleus, and cytoplasm were drawn and labelled using 10x
or 40x objective lens.
9. The waste was disposed in the bin provided and the apparatus was
cleaned.
Results and observation:

Parenchyma:
The primary cell wall is very thin and flexible. The secondary cell wall is
absent. There is a present of large vacuole at the centre of the cell. There is a
present of gap between the cell called intercellular cell. The cell are alive at
maturity and retain the ability to divide. The distribution of parenchyma is at
the large centre vacuole. The presence of intercellular space enables the
tissue to repair wound.
Collenchyma:
Conclusion:
The objective at the experiment were achieved. Cytoplasm, cell wall and
nucleus can be observed. Cytoplasm, cell wall and nucleus can be observed
by using 10x and 40x objective lens. Cell wall has a fixed shape. This is due
to the present of cellulose.
