5. OPERATING PRINCIPLE
The working area is ventilated by an unidirectional and recirculated air flow (1), whose rate is
about 70% of the total air volume, while the remaining 30% is exhausted (2) from the cabinet
via an output located on the top. The exhausted airflow draws an equal inward air flow volume
(3) through the front aperture that creates a barrier of protection for the operator. This air flow
barrier (contaminated from the outside) is suctioned (4) together with the laminar air flow
(contaminated by the product) to create 100% of the air flow (contaminated), then the cycle is
repeated.
The total air flow (100%) is supplied by a fan* blowing into a plenum from which a portion of
the air flow, after HEPA filtration, supplies the working area and the remaining, also after HEPA
filtration, is exhausted from the cabinet.
*NOTE: in the size 1.8 two fans are fitted.
HEPA filtration assures both the air sterility of the working space (product protection) and the
air sterility of the exhausted air (environment protection).

6.3 UTILITIES

6.3.1 Gas lines

WARNING! See ASSESSMENT OF RISKS, warnings 1.1.11 and 1.1.12

Inlet hoses for the combustible gas and for inert fluids and vacuum connections are
located on the rear of the right-hand compartment top (Pic. 11) and marked with
suitable labels (Pic. 13 and Pic. 14).

Pic. 11 Pic. 12

Pic. 13 Pic. 14

The taps are located on the right sidewall of the inside working chamber (Pic. 12).
The colour of the combustible gas tap knob is yellow.
The colour of the inert fluids and vacuum tap knob is black.
Combustible gas
Combustible gas line is controlled by a normal closed solenoid valve and a ball valve tap in
line, and it should be connected according to local regulations.

The solenoid valve is activated in MODE 1 only by the key of the keypad.
Alarm condition will deactivate the function.

Gas supply is automatically cut off by the solenoid valve in case of sudden lack of the vertical
airflow or in case of alarm activation (EN 12469, annex A.9). This is a safety measure to
prevent HEPA filter damage and possible risk of fire. Termination of alarm conditions and
automatic restart of the cabinet after black out will not reactivate the valve.

NOTE 1: the provision of gas supply is not recommended. Flames from gas burners interfere
with laminar airflow within the cabinet and may compromise containment.
NOTE 2: where practicable, disposable loops or use of electrical loop sterilizers are preferred
to gas burners.

Vacuum and inert fluids

Non combustible pressurised fluids line is equipped with a ball valve tap. Fluids must be non
aggressive towards materials of which the valve is made (body in brass, ball in chrome-
plated brass, gaskets in PTFE).

are fitted inside the light box, below the access panel of the filter compartment, out
of the working area.

Always enabled, activated by the key of the keypad.

Interlocked with UV-C light.

6.3.7 UVC germicidal lamp

WARNING! See ASSESSMENT OF RISKS, warnings from 1.1.6 to 1.1.8

UVC lamp is fitted inside the working chamber (Pic. 18) on the back wall.
On request is available also an optional MOBILE KIT to connect to the dedicated
socket and to lay on the working surface (Pic. 19).
Both methods assure disinfection of the cabinet working area.
After the cleaning of working area surfaces was done, it is possible to sterilize them
by using UV-C (germicidal) radiations.
NOTE 1: not suitable in case of access to contaminated area or components (i.e. for
maintenance, replacement of filters or fan), before routine control and every time big
spillage of contaminated liquids occurs.
These cases require a complete and effective decontamination of the cabinet (see
8.2).
Generally the effectiveness of UV-C lamp disinfection (wavelength 253,7 nm)
depends on exposure time and on the nature of microorganisms: special tables
provide this information.
NOTE 2: ultraviolet (UV) radiation is not recommended for use in MSCs (see EN
12469, annex A.2).

Pic. 18 Pic. 19

Enabled in MODE 2 only, activated/programmed by the key of the

keypad (see 6.2.2).
Interlocked with fluorescent light.
By mean of suitable limit switches it is not allowed to light the lamp UV-C if the front
window or panel closing the front access is not fully closed.
The front glass window has a shielding capability higher than 98% of UV-C radiation.

6.4 FRONT APERTURE

6.4.1 Glass window

WARNING! See ASSESSMENT OF RISKS, warnings from 1.1.6 to 1.1.7

The front window is made of laminated safety glass and it’s part of enclosure being a
protection against the UV-C radiations for the operator, determining moreover with
its position and integrity the inflow air barrier protection.
The breakage of the window compromises both the UVC radiations protection and the
air barrier containment.
The front window is tilting and equipped with gas springs in order to be easily
opened by hand, to ensure adequate support when is open and to guarantee a firmly
closed position.
During operating in MODE 1 (see 6.7.2), any position of the window except the
working position is UNSAFE and it will turn on the alarm devices.
During operating in MODE 2 (see 6.7.4), any position of the window except the
working position will disable the UV-C function.

6.4.2 Frontal closure panel

The closure panel (Pic. 20) allows you to completely close the front access of the
cabin. It is equipped with two key locks that lock the panel to prevent the opening by
unauthorized personnel.

Pic. 20
It can be used in the following conditions:
cabinet off (protection against dust and external contamination);
cabinet in MODE 2 (UV-C cycle or decontamination).
During operating in MODE 2 (see 6.7.4), the removal of the panel closing the front
access will disable the UV-C function.

7. GENERAL RULES – GUIDELINES

WARNING! See ASSESSMENT OF RISKS, warnings from 1.1.1 to 1.1.13

In order to achieve better comfort for operators, EUROCLONE suggests the following
recommendations:

7.1 FEATURES OF LOCATION:

• Chair should assure fully support of the body;
• If the chair has adjustable low back support, match the chair backrest to support the
lower part of the spine;
• If different operators may use the cabinet, don’t assume the settings are properly set for
everybody;
• Vary the leg position frequently and avoid placing objects that limit the legroom under
the cabinet.

7.2 WORKING POSTURE:

• Keep forearms, wrists and hands aligned in a straight neutral position: avoid bending or
angling the wrist;
• Working intensely for long time poses risks: get up from the chair periodically and take
brief walks.

7.3 WORKING PROCEDURES:

• Operate the cabinet according to what prescribed by its definition (see 2.1, 2.2 and 2.3).
• DO NOT use the working area for storage of material.
• Request an efficiency test report of the cabinet after installation, filters and other
components replacement, any repositioning or dislocation, and in any case every six
months.
• Carry out the work according to a specific programme in which working steps, needed
materials and accessories (to be put in the cabinet) are listed.
• Put inside the cabinet only essential materials and equipment, before operating the
cabinet, so that nothing would pass through the front aperture during operation.
• Verify that there are no obstructions to the exhaust air outlet before turning the cabinet
on.
• Wait at least ten minutes of SAFE OPERATING condition (see 6.7) of the cabinet before
starting any operation, in order to allow the removal of the airborne contamination.
• Wash hands and arms always with a germicidal soap before and after work.
• Wear suitable clothing, long sleeves gowns with elastic cuffs, and rubber gloves.
• Put objects at least 15 cm off from the front window and operate in the innermost part of
the safe working area (see 6.5.1).
• Avoid that contaminated material passes over the sterile one when shifting it; keep
contaminated material separate from the sterile one, place the latter far from the
working area and put the wastes in a back corner of the working surface.
• Avoid obstructing the slots of the front intake grill with sleeves of the working garment;

ORIGINAL INSTRUCTION

• DO NOT work when the fan is off (MODE 2), when audible or visible alarms are
activated, when the display does not work, when the front window is out of the working
position.
• Limit the activities inside the room where the cabinet is placed, because they could
disturb the air barrier containment and the laminar flow (LAF), with consequent decrease
in the operator and product protection.
NOTE: each time doors of the laboratory are open and closed, drafts are produced. It is
consequently advisable to put an advice board such as:
“Safety cabinet on – no entrance”.
• Keep the cabinet on for at least 10 minutes once the work is over, in order to remove
any contaminating substance from inside.
• Repeat the decontamination of inside surfaces once all objects as materials and
equipment have been taken out. For cleansing and decontamination, see par. 8.
• Pay special attention when removing cultures that have been accidentally poured inside
the working area: they could actually favour the growth of fungi and the spreading of
their spores with consequent contamination of the cabinet.

8. CLEANSING AND DECONTAMINATION

8.1 CLEANSING

8.1.1 Painted surfaces

• Use common detergents, like soap solutions.
• DO NOT USE solvents or other paint aggressive products.
8.1.2 Stainless steel surfaces
• Specific products are recommended for normal cleaning.
• The disinfection of the inner surfaces of the working space shall be carried
out by using disinfectant liquids, following the relating instructions to prevent
misuse.
Decontaminate by swabbing with a disinfectant chlorine-free product.

WARNING! r
• Chlorine products damage stainless steel surfaces.

• DO NOT use products containing chlorine for stainless steel surfaces.

8.1.3 Front glass windows

• Disinfection of the glass window should be carried out with isopropyl alcohol.

WARNING! See ASSESSMENT OF RISKS, warnings 1.1.9 and 1.1.10

8.2 DECONTAMINATION
Routine decontamination of the working area surfaces can be carried out by means of UVC
radiation (see 6.3.7).
Otherwise, in several cases is required a complete and effective decontamination of the cabinet.
This type of decontamination of the cabinet is mandatory in the following cases:

before operations requiring access to any potentially contaminated area;

every time big spillage of contaminated liquids occurs;
before maintenance (fan and filters replacement);
before routine control;
before dislocation;
after any emergency.

Users shall identify the decontaminating method effective against the agent in question.
• A method of decontamination is described in the PART 2 – Service and maintenance.
